丹皮酚对非酒精性脂肪肝大鼠炎症因子表达的影响

摘    要:目的研究丹皮酚对非酒精性脂肪肝(NAFLD)大鼠炎症因子白介素2(IL-2)、白介素6(IL-6)、肿瘤坏死因子α(TNF-α)及核转录因子κB(NF-κB)表达的影响。方法将高脂饮食诱导的NAFLD大鼠随机分成模型对照组,阳性对照组,丹皮酚高、中、低剂量组,另设正常组。观察大鼠造模及用药期间的活动情况;显微镜观察肝脏病理学变化;ELISA法检测大鼠血清中IL-2、IL-6、TNF-α和NF-κB的含量;qRT-PCR法检测肝脏中炎症因子IL-2、IL-6、TNF-α和NF-κB的基因表达量。结果正常组大鼠肝细胞无脂肪变性,模型组大鼠肝细胞存在显著脂肪变性,丹皮酚各剂量组肝细胞脂肪变性减轻,仅见到少量脂滴空泡。丹皮酚各剂量组血清IL-2、IL-6、TNF-α、NF-κB的含量及其在肝组织中mRNA的表达量均比模型组明显下降(P<0.05),呈一定的量效关系。结论丹皮酚可能通过下调大鼠炎症因子IL-2、IL-6、TNF-α以及NF-κB的表达以治疗高脂饮食诱导的NAFLD。

Abstract:Objective To study the effect of paeonol on the expression of inflammatory factors interleukin 2 (IL-2), interleukin-6 (IL-6),tumor necrosis factor (TNF-α) and transcription factors influencing B (NF- κB) in rats with non-alcoholic fatty liver disease (NAFLD). Methods NAFLD rats induced by high-fat diet were randomly divided into model control group,positive control group,paeonol high,medium and low dose groups,and normal group. The activity of rats during modeling and medication was observed. The pathological changes of liver were observed by microscope. The contents of IL-2,IL-6,TNF-α and NF-κB in serum were detected by ELISA. The gene expression levels of IL-2,IL-6,TNF-α and NF-κB in liver were detected by qRT-PCR. Results No fatty degeneration occurred in the liver cells of the normal group. Significant fatty degeneration was observed in the hepatocytes of the model group. The hepatic steatosis of the paeonol doses groups was alleviated,and only a small amount of lipid droplet vacuoles were observed. The serum levels of IL- 2, IL- 6, TNF- α, NF- κB and the expression of mRNAs in liver tissue were significantly lower than those in the model group,which showed a doseeffect relationship. Conclusion Paeonol may down-regulate the expression of inflammatory cytokines IL-2,IL-6, TNF-α and NF-κB in rats to treat NAFLD induced by high-fat diet.
