细胞壁缺陷沙门菌细胞多糖的研究

目的探讨细胞壁缺陷沙门菌的细胞多糖组成特点及其变异的生化机制。方法采用分光光度法、鲎血细胞溶解物试验,分别对伤寒沙门菌和甲型副伤寒沙门菌的细胞壁缺陷突变株(CWDM)以及伤寒沙门菌的CWDM返祖菌株的细胞壁多糖、细胞外多糖、脂多糖进行定性、定量实验,检测与分析沙门菌CWDM及其返祖菌株细胞多糖的化学组成特点。结果伤寒沙门菌及甲型副伤寒沙门菌的CWDMs细胞壁多糖与细胞外多糖的含量明显低于其亲代细菌型和伤寒沙门菌粗糙型返祖菌,鲎血细胞溶解物试验定性试验结果显示,两种细菌的CWDMs均表现为阴性反应,但定量试验表明其含量仅为细菌型和伤寒沙门菌返祖菌含量的1/30。结论细胞壁缺陷可导致沙门菌发生细胞壁多糖、细胞外多糖及脂多糖的含量减少。

Study on Polysaccharide of Cell Wall Deficient Salmonella. Spp

Objective To determine the quantity and composing of the polysaccharide of the cell wall deficient Salmonella spp.and the mechanism of the variation.Methods The quantity of polysaccharide of cell wall and outer-cell of Salmonella typhi(St),Salmonella paratyphy A(SpA),the cell wall-deficient mutants(CWDMs) derived from the Salmonella typhi and Salmonella paratyphy A and the rough strains of S.typhi reverted from the CWDM of S.typhi were determined by the classical biochemical methods.The amount and activities of lipopolysaccharides(LPS) from the CWDMs,the bacterial forms and the rough strains of S.typhi were determined by limulus amoebocyte lysate test(LALT).Results The quantity of the polysaccharide of cell wall,outercell,the amount and activity of the LPS were remarkably lower than the parental bacteria and the rough strains of S.typhi.Conclusion The cell wall deficiency would lead to the change of polysaccharide of cell wall and outer-cell of Salmonella spp in quantity and to low the quantity and activity of LPS.
