自体骨髓间充质干细胞心肌移植后的分化及存活

摘    要:背景:骨髓间充质干细胞移植入急性心肌梗死组织后是否能迁移、分化及存活,目前尚不清楚.目的:观察兔骨髓间充质干细胞移植入急性心肌梗死组织后,在体内的迁移、分化及其存活.方法:分离培养兔骨髓间充质干细胞,体外DAPI标记细胞.结扎兔左前降支制作急性心肌梗死模型,造模成功后30只新西兰大耳兔抽签法分为骨髓间充质干细胞组和对照组,每组15只.骨髓间充质干细胞组于冠状动脉结扎后1 h在梗死周边区用微量注射器分4点注射自体骨髓间充质干细胞.对照组于相同部位注射等量生理盐水.每点注射体积30 μL.分别于移植后10 min,3 d,4周时各组各处死5只动物.取心脏行冰冻切片,荧光显微镜观察DAPI标记骨髓间充质干细胞的分布情况;免疫荧光法检测标记细胞是否表达心肌特异性肌钙蛋白Ⅰ和α-肌动蛋白.结果与结论:DAPI标记的骨髓间充质干细胞在兔急性心肌梗死模型心脏微环境中,蓝色细胞核分布广泛,呈椭圆形,似心肌细胞核,其排列方向与宿主心肌纤维方向一致,间接免疫荧光法检测胞浆心肌特异性蛋白肌钙蛋白Ⅰ、α-肌动蛋白均为阳性.说明缺血心肌内移植入骨髓间充质干细胞,可在局部微环境的刺激下向心肌细胞定向分化.
