摘    要:用递增剂量的吗啡处理 SD大鼠 ,建立吗啡依赖大鼠模型 ,选取中脑导水管周围灰质、伏核、纹状体等核团 ,提取总 RNA,用抑制性消减杂交 ( SSH)方法 ,分离吗啡依赖大鼠的高表达基因 ,初步获得了 4个新基因的 c DNA片段 ,并发现了 3个已知的功能基因在吗啡依赖过程中增高。进一步的研究尚在进行中。

Abstract:The programmed increasing doses of morphine were given to SD rats to establish morphine dependent model. Brain areas of periqueductal gray(PAG). nucleus accumbens and striatum were dissected for total RNA extraction. Suppression subtractive hybridization (SSH) method was differentially used for isolating expressed cDNA fragments of morphine dependent rats. Eight highly expressed cDNA fragements were obtained from morphine dependent rat brains, including four new cDNA fragements and 3 known genes. Further studies are still in progress to characterized the full length gene and clarify their functions in morphine dependence.
