摘    要:本文应用基因工程技术,设计并化学合成了编码恶性疟原虫红内期保护性抗原MSA_1,MSA_2和RESA上多个免疫原性决定簇(含T、B细胞位点)以及来自白细胞介素-1(IL-1)和破伤风类毒素(TT)上的外源性T细胞激活位点的复合基因HGFC。全基因共由246个碱基对组成,编码一个75肽的复合抗原。合成后的基因克隆在M13mp18载体上,并经序列分析证实与设计完全一致。杂合基因合成和克隆的成功,为在基因水平上进行多价疟疾疫苗的蛋白质工程研究创造了条件。

Abstract:A hybrid gene named HGFC coding for several protective epitopes of MSA1,MSA2 and RE-SA in the erythrocytic stages of Plasmodium falciparum as well as exogenous T cell enhancer epitopes from interleukin-1(IL-1) and tetanus toxoid(TT) was designed and chemically synthesized. The gene was composed of 246 base pairs and coded for a hybrid antigen of 75 amino acids. This synthesized gene was cloned into the M1 3mpl8 vector and was proved fully correct just as designed by hybridization, restriction analysis and DNA sequencing. This hybrid gene was a useful material for protein engineering researches of polyvalent malaria vaccine at the gene level.
