痴呆模型小鼠脑神经组织生化工程指标的改变及乌圆补血口服液对其重建的影响

摘    要:目的研究老年痴呆(AD)小鼠模型脑神经组织生化工程指标的改变及乌圆补血口服液对其重建的影响。方法将小鼠分成模型组、治疗组和正常组,模型组和治疗组小鼠腹腔注射D-半乳糖和亚硝酸钠混合液,每天1次,连续60 d造模。造模30 d后,治疗组用乌圆补血口服液灌胃治疗,模型组和正常组用等体积蒸馏水灌胃。y型水迷宫测定小鼠治疗前后记忆功能。实验结束,处死小鼠,取大脑制备10%脑匀浆,测定脑匀浆生化指标。结果脑匀浆乙酰胆碱酯酶(TchE)活力模型组、正常组和治疗组分别为(1.709±0.498)IU/mg、(1.436±0.313)IU/mg、(1.182±0.157)IU/mg,模型组明显高于其他2组(P<0.05和P<0.01);丙二醛(MDA)含量分别为(7.935±0.775)μmol/L、(6.934±1.284)μmol/L、(6.356±0.799)μmol/L,模型组明显高于其他2组(P<0.05);脑(O-.2)氧自由基清除率分别为(2.346±1.167)%、(7.366±3.413)%、(8.672±3.519)%,治疗组和正常组均明显高于模型组(P均<0.01);实验前各组水迷宫错误率分别是9.91%,7.41%,14.81%;实验结束时错误率分别是10.10%,1.23%,1.39%,实验结束时模型组水迷宫错误率明显高于治疗组和正常组;其他生化指标脑蛋白含量、脑GSH-PX等各组间无显著性差异。结论 D-半乳糖和亚硝酸钠联合造痴呆模型小鼠大脑神经组织胆碱酯酶活性等生化指标有明显改变,乌圆补血口服液对其重建有明显的促进作用。

Abstract:Objective It is to study the level changes of biochemical engineering in cerebral nerve organization of dementia model mouse and the influence of Wuyuan Buxue oral liquid on its reconstruction.Methods The mice were divided into model group,treatment group and normal group with D-galactose and sodium nitrite mixture to the model group and treatment groups of mice by intraperitoneal injection,once a day,continuously molding 60 days.After model for 30 days,the treatment group was treated with Wuyuan Buxue oral liquid gavage,the model group and normal group were treated the equal-volume of distilled water orally.With y-type water maze in mice measured before and after treatment of memory function.The end of the experiment,mice were sacrificed,the brain was taken 10% brain homogenate preparation,determination of biochemical indicators of brain homogenate.Results Brain homogenate acetylcholinesterase(TchE) dynamic in model group,normal group and treatment group were(1.709±0.498) IU/mg,(1.436±0.313) IU/mg and(1.182 ± 0.157) IU/mg respectively,which in model group was significantly higher than that in the other two groups(P<0.05 and P<0.01).Malondialdehyde(MDA) contents were(7.935±0.775) μmol/L,(6.934 ± 1.284) μmol/L and(6.356 ± 0.799)μmol/L respectively,which in model group was significantly higher than that in treatment group(P<0.05).Brain superoxide anion radical() scavenging rate were(2.346±1.167)%,(7.36±3.413)% and(8.672±3.519)% respectively,which in treatment group and normal group was significantly higher than that in model group(P<0.01).In each group the experiment error rate of water maze before experiment was 9.91%,7.41% and 14.81% respectively and at the end of experiment the error rate was 10.10%,1.23% and 1.39% respectively,which in model group at the end of experiment was significantly higher than that in treatment group and normal group.There was no significant difference on other biochemical indexes such as brain protein content,brain GSH-PX and so on among those groups.Conclusion Brain cholinesterase activity in organizations such as biochemical markers in dementia model mouse made with D-galactose united with sodium nitrite have obvious changes and Wuyuan Buxue oral liquid has obvious promotion action on its reconstruction.
