摘    要:目的:确定杜氏利什曼原虫39KD抗原基因的基因分析与同源性。方法:以杜氏利什曼原虫39KD抗原基因为探针,与不同种株的利什曼原虫进行Southem杂交,确定不同种株的利什曼原虫该基因的变化,并将该基因序列测定后,进行DNA序列数据库检索,分析其同源性,并进行数据库登记。结果:杜氏利什曼原虫不同地域株及婴儿利什曼原虫基因组中均含有该基因,并且无明显定位变化,整个基因在DNA数据库中无完全DNA同源序列,在GenBanK中的序列号为3280。结论:该抗原基因是利什曼原虫保守基因。

Abstract:We have previously cloned and expressed a DNA fragmnt encoding 39KD antigen of Leishmania donovani promastigotes using the λgtll DNA as the vector, the expression product was not fused to the β-galactosdase that encoded by the lead sequence of the vector. In this paper, we sequenced the 39KD antjgen gene and demonstrated the DNA sequence homologies using the FASTAN and the BLASTN algorithm. Result that the 5' terminal sequence similar to the interval region of the α-tubulin and the gGADPH, and similar to the promoter sequence from prokaryotic cell. Then, the genomic DNA of different Leishmanis species and strains was identified to the 39KD antigen gene using Southem blot technique. Result that the 39KD antigen gene distributed in all Leishmania species and strains. These shown the 39KD antigen gene is a specific gene of genus Leishmania., and the 5' terminal seuence of the 39KD antigen gene can Promote gen expressing in Leishmania and prokaryotic cell.
