血清和糖皮质激素诱导的蛋白激酶1在梗阻性肾病肾小球中的表达及姜黄素对其的影响

摘    要:目的:探索在单侧输尿管梗阻(UUO)性肾病变进程中,血清和糖皮质激素诱导的蛋白激酶1(SGK1)在肾小球中的表达变化及姜黄素对其的影响。方法:采用右侧输尿管接扎术诱导大鼠UUO模型,将54只大鼠随机分为假手术组、模型组、姜黄素组3组,每组18只。术后第3、7及14 d分别随机处死各组中的6只大鼠,检测血尿素氮(BUN)、血清肌酐(SCr)。应用免疫组化技术检测肾小球中SGK1蛋白的表达。结果:模型组大鼠肾小球中的SGK1蛋白的表达第3 d开始上调,第7 d和14 d继续升高。与模型组相比,姜黄素组BUN、SCr的改变不明显,但肾小球中的SGK1蛋白表达均显著下降。结论:随UUO肾病变发展,肾小球中SGK1持续高表达,进一步提示SGK1在梗阻性肾病的肾小球纤维化病变进程中起重要作用。姜黄素能抑制肾小球中SGK1的表达,从而抑制肾小球的纤维化。

Abstract:Objective: To investigate the dynamic expression of serum-and glucocorticoid-induced kinase 1(SGK1) in the renal glomeruli of nephropathy patients with unilateral urethral obstruction(UUO) and its response to Curcumin.Methods: Fifty-four Sprague-Dawley rats were randomly divided into a sham operation,a UUO model and a Curcumin treatment group of 18 each.Six rats of each group were killed respectively at 3,7 and 14 days after surgery,the levels of blood urea nitrogen(BUN) and serum creatinine(SCr) observed,and the expression of the renal glomerular SGK1 protein detected by immunohistochemistry. Results: The renal glomerular SGK1 was up-regulated in the UUO model group from the 3rd to the 14th day.While in the Curcumin treatment group,a significant decrease was observed in the expression of renal glomerular SGK1,but with no obvious change in the level of BUN and SCr.Conclusion: The expression of renal glomerular SGK1 remains high with the progression of UUO,which suggests that SGK1 may play an important role in the development of obstructive nephropathy.Curcumin could prevent renal glomerular fibrosis by inhibiting the expression of SGK1.
