急性肺缺血再灌注引起肺组织表面活性物质相关蛋白A变化实验研究

摘    要:目的:探讨肺急性肺缺血再灌注(ischemia reperfution,I/R)时表面活性物质相关蛋白A(surfactant-associated protein A,SP-A)含量的变化与肺损伤的关系。方法:40只健康的SD大鼠,雌雄不拘,随机分成5组:假手术组(S)、单纯缺组(I)、再灌注1h组(R1组)、再灌注2h组(R2组)、再灌注6h组(R3组)。根据Eppinger模型建立原位阻断左肺门大鼠温肺I/R模型。术毕测定各组血清丙二醛(MDA)及肺湿/干重比例(W/D)、病理观察、用Western-Blot及免疫组化法检测各组肺组织中SP-A。结果:(1)MDA、肺W/D在I组即较S组开始升高,予再灌注后,各组MDA、肺W/D随再灌注时间的延长明显升高(P<0.05);(2)SP-A在S组表达最多,在I组表达减少,再灌注后随时间的延长表达更少(P<0.05)。结论:大鼠肺急性I/R后,肺表面活性物质相关蛋白A减少是造成肺一系列损害的重要因素。
