胰岛素样生长因子对新生大鼠缺氧缺血性脑损伤的干预作用

摘    要:目的:研究外源性胰岛素样生长因子-1(insulin like growth factor-1,IGF-1)对缺氧缺血新生大鼠脑组织中超氧化物歧化酶(SOD)和一氧化氮(NO)水平的影响,探讨IGF-1治疗新生大鼠缺氧缺血性脑损伤(HIBD)的作用机制。方法:将7日龄wistar大鼠75只随机分为:①正常组(n=15):不干预。②HIBD模型组(n=15):麻醉后结扎左侧颈总动脉,1 h后置于37℃密闭容器中,给予吸0.08氧气,持续2 h,制备脑组织缺氧缺血模型。③生理盐水对照组(n=15):制备模型同前,模型成功后腹腔注射生理盐水(15 ml/kg)。④丹参干预组(n=15):模型成功后腹腔注射丹参注射液(1.5 g/kg)。⑤胰岛素样生长因子干预组(n=15):制备HIBD模型后腹腔注射IGF-1(1.5 ug/kg)。缺氧缺血后72 h处死,取大脑皮质,制作脑组织匀浆,检测脑组织中SOD、NO水平。结果:HIBD组与NS组脑组织中SOD水平较正常组低,HIBD组与NS组脑组织中NO水平较正常组高,同正常组比较有统计学差异(P<0.01),IGF-1组与丹参治疗组SOD水平较模型组高,IGF-1组与丹参治疗组NO水平较HIBD模型组低,同模型组比较有统计学差异(P<0.01)。结论:IGF-1可促进大脑皮质中SOD水平增高,NO水平降低,对HIBD有防治作用。

Abstract:Objective:To study the effect of exogenous insulin-like growth factor-1(IGF-1) on the levels of superoxide dismutase(SOD) and nitric oxide(NO) in brain tissues of neonatal rats with hypoxic ischemic brain damage(HIBD),explore the mechanism of IGF-1 in treatment of HIBD.Methods:75 7-day old Wistar rats were divided into different groups randomly: ①normal group(15 rats): without intervention;②HIBD model group(15 rats): left arteria carotis communis was ligated after anesthesia,then the rats were put into a 37℃ closed container after one hour,the rats were treated with inhalation of 0.08 oxygen of air for 2 hours,HIBD models were prepared;③normal saline control group(15 rats): the method to prepare HIBD models was similar to HIBD model group,then the rats were treated with intraperitoneal injection of normal saline(15 ml/kg);④salvia intervention group(15 rats): the method to prepare HIBD models was similar to HIBD model group,then the rats were treated with intraperitoneal injection of salvia injection(1.5 g/kg);⑤IGF-1 intervention group(15 rats): the method to prepare HIBD models was similar to HIBD model group,then the rats were treated with intraperitoneal injection of IGF-1(1.5 μg/kg).The rats were killed at 72 hours after hypoxia and ischemia,the cerebral cortex was obtained to prepare brain tissue homogenate;the levels of SOD and NO in brain tissues were detected.Results:The levels of SOD in HIBD model group and normal saline control group were lower than that in normal group,the levels of NO in HIBD model group and normal saline control group were higher than that in normal group,there was significant difference(P<0.01);the levels of SOD in IGF-1 intervention group and salvia intervention group were higher than that in HIBD model group,the levels of NO in IGF-1 intervention group and salvia intervention group were lower than that in HIBD model group,there was significant difference(P<0.01).Conclusion:IGF-1 can promote the increase of SOD level and the decrease of NO in cerebral cortex,which can prevent and treat HIBD.
