摘 要: | 嵌合免疫受体是一类在体外通过基因重组方法而建立的一种能识别特异性靶抗原的免疫受体。运用从噬菌体表面展示技术筛选出的能与GRP78高亲和力结合的靶向肽段做为抗原识别段,同时将膜结合型IgG3蛋白的Hinge-CH2-CH3做为连接穿膜段,并首次将FcαRa.1(CD89)分子的胞内部分做为胞内信号转导段组建了四种新型免疫嵌合受体,将其中一种嵌合受体转入U937细胞系,并使其获得了部分识别杀伤其他肿瘤细胞的能力。

Construction of four different novel chimeric immune receptors |

Abstract: | Chimeric immune receptors(CIR) are immune receptors of recognizing specific antigens by means of gene recombination construction.We reported here for the first time that 4 different novel chimeric immune receptors were constructed successfully,which consist of GRP78 targeted peptide resulted from affinity panning of libraries of bacteriophages,mIgG3 hinge-CH2/CH3 constant domain and a transmembrane and cytoplasmic region of FcαRa.1(CD89) to induce cellular activation.One of the CIRs recognizing GRP78 was engrafted into U937 via retrovirus and U937 cells consequently obtained the ability of lysing tumor cells. |
