摘    要:目的建立同时测定板栗种仁中原儿茶酸、对羟基苯甲酸、异香草酸含量的方法。方法采用Dikma Diamonsil C18柱(250 mm×4.6 mm,5μm),以甲醇-体积分数为0.05%的甲酸水溶液为流动相进行梯度洗脱,流速为1.0 mL.min-1,检测波长为260 nm,柱温为室温。结果原儿茶酸?对羟基苯甲酸、异香草酸质量浓度分别在10~100 mg.L-1?5~50 mg.L-1?50~500 mg.L-1内与峰面积具有良好的线性关系(r≥0.999 8,n=6),方法平均回收率分别为98.5%?98.0%?99.6%。结论本方法可用于同时测定板栗种仁中原儿茶酸?对羟基苯甲酸、异香草酸的含量。

Abstract:Objective To develop an RP-HPLC method for the simultaneous determination of the contents of protocatechuic acid, p-hydroxybenzoic acid and isovallic acid from seeds of Castanea mollissima Blume. Methods The analysis was carried out on a Dikma Diamonsil C18 (250 mm×4.6 mm, 5 mm) column; the mobile phase was composed of methanol and 0.05% formic acid with gradient elution at a flow rate of 1.0 mL·min-1. The detection wavelength was set at 254 nm and column temperature was room temperature. Results The linear ranges of protocatechuic acid, p-hydroxybenzoic acid and isovallic acid were 10-100 mg·L-1, 5-50 mg·L-1, 50-500 mg·L-1r≥0.999 8, n = 6), respectively. The average recoveries were 98.5%, 98.0%, 99.6%, respectively. Conclusion This method is simple and can be used to determine the contents of the above three components with satisfactory accuracy and reproducibility.
