脑缺血大鼠HGF及VEGF的表达及其相关性

摘    要:目的研究脑缺血大鼠肝细胞生长因子(HGF)及血管内皮细胞生长因子(VEGF)的表达情况及其相关性。方法48只雄性Wistar大鼠随机分为假手术组及大脑中动脉线栓法制作局灶性脑缺血大鼠模型组,分别于2h、6h、12h、24h、3d、7d、14d处死动物,采用免疫组织化学法观察各组脑组织HGF和VEGF的表达。结果HGF在假手术组几乎无表达,缺血2hHGF表达增加,并随时间延长表达逐渐上升,缺血组与对照组比较均有统计学差异(P〈0.01)。VEGF假手术组即有表达,缺血6h表达明显增加,在3-14d时表达增高达高峰,缺血组与对照组比较均有统计学差异(P〈0.01)。结论脑缺血后HGF及VEGF表达增高,两者可能起相互协同促进作用,共同参与缺血灶周围血管新生及脑保护作用。

Abstract:Objective To determine the expression of hepatocyte growth factor (HGF) and vascular endothelial growth factor (VEGF) in cerebral ischernic rat and analyze their relationship. Methods There 48 male Wistar rats were randomly divided into sham operation group and the ischemic group. Cerebral ischemia models of rats were established by blocking the middle cerebral artery. The expression of HGF and VEGF in the two groups were detected by immunohistochemical method on different times of 2h、6h,12h、24h、3d、7d、14d. Results HGF in the sham-operated group was almost without expression. While the expression of HGFin ischemic group increased at 2h along with the increase of time. There significant differences were observed in expression between the ischemia group the control group (P〈0.01). VEGF was expressed in sham-operated group and markedly increased in expression in ischemia group at 6h and reached a peak during 3-14d , showing significant differences between the isehemia group and the control group (P〈0.01). Conclusion The expression of HGF and VEGF increased after cerebral ischemia,and there is a mutual effect and both of them participated in the angiogenesis and protection the brain around ischemic loci .
