稳心颗粒对缺血大鼠心室肌电生理特性的影响

摘    要:目的观察不同浓度稳心颗粒对大鼠缺血左心室肌跨膜动作电位及有效不应期等电生理特性的影响。方法将40只SD大鼠随机分为缺血对照组,含有1、5、10 g/L稳心颗粒给药组,每组10只。脱臼法处死大鼠后迅速分离左心室楔形心室肌组织块。利用标准玻璃微电极技术,记录缺血前、灌注单纯缺血液或灌注含不同浓度稳心颗粒缺血液时不同刺激周长下大鼠左室心内膜跨膜动作电位(TAP)及有效不应期(ERP)。结果①与正常对照组比较,缺血对照组左心室肌动作电位幅值(APA)降低,动作电位时程(APD)缩短,0期最大去极速度(Vmax)减小,有效不应期(ERP)缩短(P〈0.05);与缺血对照组比较,各稳心颗粒给药组APA、APD、Vmax增大(P〈0.05),5、10 g/L稳心颗粒给药组ERP明显延长(P〈0.05)。②缺血液灌流使心肌组织的频率依赖性减弱,随着稳心颗粒给药浓度的增加和给药时间的延长,心肌组织的频率依赖性特点逐渐恢复。结论稳心颗粒可减轻缺血引起的心室肌电生理改变,这可能是其抗心律失常的机制之一。

Abstract:Objective To investigate the effect of the Chinese medical compound Wenxin Granule on the electrophysiological characteristics of ventricular myocytes in ischemia-reperfusion SD rats.Methods 40 SD rat were randomly divided into ischemia-reperfusion,1,5,10 g/L Wenxin Granule respectively treatment groups.10 cases in each group.The left ventricle muscle tissue was separated immediately after rats were dislocated,and the wedge were perfused.Standard glass microelectrode technique was used to record the transmembrane action potential and effective refractory period at the different cycle length stimulation.Results ①Compared with control group,the APA,APD,Vmax were decreased and ERP were reduced significantly in ischemia-reperfusion group(P < 0.05);while the APA,APD,Vmax were significantly increased in Wenxin Granule treatment groups(P < 0.05),and the ERP prolonged significantly in 5,10 g/L Wenxin Granule respectively treatment groups compared with ischemia control group.②The characteristic of frequency dependence in cardiac muscular tissue were weaken after perfused with ischemia solution.Wenxin Granule could recover the characteristics with the increase of dosing concentration and the extension of time.Conclusion Wenxin granules could reduce instability of electrophysiological characters and plays an important role in anti-arrhythmia.Wenxin granules can recover ventricular muscles blood-oxygen supply and may improve the function of ion pump.
