羟丙基-β-环糊精对葛根素溶解性与角膜渗透性的影响

摘    要: 目的研究羟丙基-β-环糊精(HPCD)对葛根素溶解性与角膜透过性的影响。方法采用了相溶解度法和离体角膜扩散实验。结果在不同浓度HPCD下,葛根素的相溶解曲线呈AL型;随着HPCD浓度的增大,葛根素的表观渗透系数减小。50 g·L-1HPCD增溶的表观渗透系数分别是以150 g·L-1HPCD和40 g·L-1PVP增溶的3.12和2.86倍,但150 g·L-1HPCD与40 g·L-1PVP增溶的表观渗透系数相比没有显著性差异(P>0.05)。结论HPCD能增加葛根素的溶解度与提高其角膜透过性。

Abstract:OBJECTIVE To investigate the effect of hydroxypropyl beta cyclodextrin(HPCD)on aqueous solubility and corneal permeation of puerarin.METHODS The solubility of puerarin with HPCD was investigated by phase-solubility method.The permeability of puerarin across isolated rabbits cornea was measured in vitro.RESULTS An AL-type phase solubility curve was obtained.The apparent permeation coefficient of puerarin decreased as the concentration of HPCD increased.The apparent permeation coefficient at 50 g·L-1HPCD 1evel was increased by 3.12 times as that at 150 g·L-1HPCD 1evel,and 2.86 times as that at 40 g·L-1PVP 1evel.There was no significant difference(P>0.05)between the apparent permeation coefficient at 150 g·L-1HPCD 1evel and 40 g·L-1PVP 1evel.CONCLUSION The solubilizing effect of HPCD on puerarin in aqueous solution is good.HPCD enhances the permeability of puerarin across isolated rabbits cornea.
