咖啡因对实验性自身免疫性脑脊髓炎大鼠的IFN-γ和TGF-β的影响

摘    要:目的:探讨咖啡因对实验性自身免疫性脑脊髓炎(EAE)大鼠的中枢神经系统炎性因子干扰素-γ(IFN-γ)、转化生长因子-β(TGF-β)的影响。方法:Wistar大鼠随机分为EAE模型阳性对照组、咖啡因处理组(10mg/kg、30mg/kg)、阴性对照组。复制豚鼠脊髓匀浆诱导Wistar大鼠的EAE模型,应用咖啡因干预,观察EAE的发病率变化;通过HE染色技术观察神经组织炎性细胞浸润情况;应用半定量RT-PCR法测定颈髓IFN-γ、TGF-β含量。结果:咖啡因处理组大鼠EAE发病率下降、脑脊髓组织炎性细胞浸润减轻;EAE模型阳性对照组、咖啡因处理组(10mg/kg、30mg/kg)、阴性对照组的颈髓IFN-γ含量分别为1.23±0.58、0.65士0.10、0.25±0.07、0.26±0.12,TGF-β含量分别为0.91±0.10、1.05±0.01、1.19±0.01、0.75±0.07。结论:咖啡因处理可以减少IFN-γ、提高TGF-β的含量,减轻EAE的中枢神经组织损伤。

Abstract:Objective: To investigate the effect of caffeine on IFN-γ and TGF-β in the central nervous system in Wistar rat with experimental autoimmune encephalomyelitis. Methods: Rats were randomly divided into EAE positive control group, caffeine treatment groups (10 mg/kg, 30 mg/kg) and negative control group. The FAE animal model was induced on Wistar rat by the guinea pig spinal cord homogenates. We investigated the effect of caffeine treatment on the FAE incidence, the infiltration of inflammatory cells by hematoxylin-eosin staining and the expression of IFN-γ and TGF-β mRNA in cervical cord by semi-quantitative RT-PCR. Results: Treatment with caffeine significantly reduced the incidence of EAE in the rats (P〈0.05) and alleviated the infiltration of inflammatory cells in spinal cord and cerebral white matter. The concentration of IFN-γ in cervical cord in EAE positive control group, caffeine treatment groups (10 mg/kg, 30 mg/kg) and negative control group was 1.23±0.58, 0.65±0.10, 0.25±0.07, 0.26±0.12, respectively. The concentration of TGF-β in cervical cord in EAE positive control group, caffeine treatment groups (10 mg/kg, 30 mg/kg) and negative control group was 0.91±0.10, 1.05±0.01, 1.19±0.01, 0.75±0.07, respectively. Conclusion: Treatment with caffeine in EAE model can relieve the inflammatory injury of nervous tissue by decreasing the expression of IFN-γ while enhancing the expression of TGF-β .
