胃泌素对胃癌细胞增殖和COX-2表达的影响

摘    要:目的 探讨胃泌素和胃泌素受体拮抗剂丙谷胺对胃癌细胞株SGC-7901增殖和环氧化酶2(COX-2)表达的调控作用.方法 SGC-7901细胞接种于含10%胎牛血清的RPMI-1640培养液中常规培养24 h后,换无血清培养基继续培养24 h,加入胃泌素(终浓度为0.1、1、10、100 nmol/L)或胃泌素(100 nmol/L)加丙谷胺(终浓度为1、10、100、1000 μmol/L),分别培养24、48和72 h;MTT比色分析细胞增殖,Western blot检测COX-2蛋白表达.结果 胃泌素呈时间和剂量依赖性地促进SGC-7901细胞增殖,而丙谷胺抑制了胃泌素诱导的细胞增殖.胃泌素增加COX-2在SGC-7901细胞的表达,丙谷胺有效抑制胃泌素诱导的COX-2表达.结论 胃泌素通过胃泌素受体诱导COX-2表达促进胃癌细胞增殖.

Effects of gastrin on the proliferation and cyclooxygenase-2 expression in gastric cancer cells
Abstract:Objective To investigate the regulation role of gastrin and a gastrin receptor antagonist proglumide on the cell proliferation and cyclooxygenase(COX)-2 expression in a gastric cancer cell line,SGC-7901.Methods SGC-7901 cells were seeded in RPMI-1640 supplemented with 10% heat-inactivated fetal calf serum and routinely incubated for 24 h.After serum-free starvation for 24 h, the cells were cultured with either gastrin at the final concentrations of 0.1,1, 10 and 100 nmol/ L or 100 nmol/L gastrin plus proglumide at the final concentrations of 1,10,100 and 1000 μmol/L for 24, 48 and 72 h, respectively.The cell proliferation was determined by MTT colorimetric assay.Expression of COX-2 was measured with Western blot analysis.Results Gastrin increased SGC-7901 cell proliferation in a dose- and time-dependent manner, which was significantly inhibited by proglumide.Gastrin increased the expression of COX-2 in SGC-7901 cells, which was abolished by proglumide.Conclution Gastrin promotes gastric cancer cell proliferation via upregulation of COX-2 expression induced through the activation of the gastrin receptor.
