摘    要:目的 研究已酮可可碱(PTX)对重症急性胰腺炎(SAP)大鼠肺损伤的保护作用.方法 健康成年雄性SD大鼠72只,随机分为对照(C)组、实验(E)组和干预(P)组.E组和P组用5%牛磺胆酸钠逆行胰胆管注射制备SAP模型.然后,P组立即腹腔注射PTX.于制模后2、6和12 h观察胰腺和肺组织的病理学改变;ELISA法测定血清中性粒细胞趋化因子(CINC)和白细胞介素6(IL-6)水平变化;Western blot法检测肺组织核因子抑制蛋白(IκBα)的表达量.结果 E组血清CINC、IL-6水平及肺组织IκBα蛋白的表达均显著高于同时段C组(P<0.05);与E组比较,P组血清CINC、IL-6水平和肺组织IκBα蛋白表达量均明显降低(P<0.05),胰腺和肺组织病理改变减轻.结论 SAP肺损伤时,肺组织IκBα明显升高,核因子κB(NF-κB)活化,从而启动CINC、IL-6等过量表达.PTX可以抑制NF-κB/IκB信号通路的激活,对SAP肺损伤具有保护作用.

Protective effects of pentoxifylline on severe acute pancreatitis-associated lung injury in rats
Abstract:Objective To study the effects of pentoxifylline(PTX)on the severe acute pancreatitis(SAP)-associated lung injury in rats.Methods Seventy-two male SD rats were randomly divided into three groups.SAP models were established in groups of E and P by retrograde injection of 5% sodium taurocholate into biliopancreatic duct.Pentoxifylline(PTX)was injected into abdominal cavity of rats in group P as soon as the model was developed.The rats iri group C were taken as the controls.ELISA was used for detecting serum levels of cytokine-induced neutrophil chemoattractant (CINC)and interleukin-6(IL-6)and Western blot analysis was used for detecting the expression of IκBα in the lung tissues at 2,6 and 12 h after model establishcment.The pathological changes of the lungs were observed as well.Results The expression of IκBα in lung tissues and serum CINC and IL-6 were higher in group E than those in groups C and P(P<0.05).The pathological changes of the lungs were lighter than those in group E(P<0.05).Conclusion The expression of IκBα in lung tissues is increased and nuclear factor kappa B(NF-κB)is activated,which lead to overexpressions of CINC and IL-6 in serum.PTX has a protective effect on the lung injury induced by SAP via inhibiting NF-κB/IκB signal pathways.
