摘 要: | 目的 探讨糖基化终产物(AGEs)对小鼠成骨样细胞基质金属蛋白酶2(MMP-2)分泌的影响.方法 用不同浓度的AGEs(50、100、200、400 mg/L)干预培养的小鼠成骨样细胞(MC3T3-E1)24 h和用200 mg/L的AGEs干预MC3T3-E1不同时间(12、24、48 h),以无血清培养基(α-MEM)和相应浓度的牛血清白蛋白(BSA)为对照.用Western blot法检测小鼠成骨样细胞上清中MMP-2蛋白的表达.结果 不同浓度和不同时间的AGEs干预的MMP-2分泌水平明显高于对照组(P<0.01).结论 AGEs以时间及浓度依赖的方式增加小鼠成骨样细胞MMP-2分泌,AGEs可能通过调节成骨细胞MMP-2的分泌参与骨质疏松的发病.

Effect of advanced glycation end products on secretion of MMP-2 in cultured mouse embryo/fetus calvaria fibroblusts |

Abstract: | Objective To investigate the effects of advanced glycation end products(AGEs)on the secretion of matrix metalloproteinase-2(MMP-2)in cultured mouse embryo/fetus calvaria fibroblasts(MC3T3-E1).Methods The cultured MC3T3-E1 was intervened with AGE-BSA(50,100,200,400 mg/L)for 24 h and with 200 mg/L AGE-BSA for 12,24,48 h,respectively,and a-MEM and BSA were taken as the negative control.The expression level of MMP-2 in the supematant of MC3T3-E1 was detected by Western blot.Results The content of MMP-2 was significantly higher in the cells intervened with different concentrations AGE-BSA or with 200 mg/L AGE-BSA for different times(P<0.01).Conclusion The AGE-BSA can stimulate the secretion of MMP-2 in cultured MC3T3-E1 in a dose-and time-dependent manner. |
