AML病人MN1和PTEN基因表达及临床意义

摘    要:目的探讨脑膜瘤1(MN1)基因和PTEN基因表达与急性髓系白血病(AML)病情发展及预后的关系。方法采用RT-PCR技术检测38例AML病人(初治组16例,缓解组12例,复发组10例)骨髓单个核细胞中MN1和PTEN mRNA的表达水平,另取非恶性血液病病人及正常人骨髓共13例作正常对照组。结果 MN1和PTEN mRNA在AML病人骨髓单个核细胞中阳性表达率分别为76.3%和60.5%。初治组MN1 mRNA表达水平较正常对照组显著升高,PTEN mRNA表达水平较正常对照组显著下降(F=2.385、2.054,q=2.305、2.867,P〈0.01);缓解组MN1 mRNA表达水平下降,PTEN mRNA表达水平上升,与初治组比较差异均有统计学意义(q=2.121、2.756,P〈0.05);在复发组中MN1 mRNA的表达水平复又升高,PTEN mRNA的表达水平复又下降,与正常对照组比较,差异均有统计学意义(q=2.361、2.723,P〈0.05)。MN1和PTEN基因在AML中的表达水平呈负相关(r=-0.321,P〈0.05)。结论 MN1和PTEN基因的表达与AML发病及预后密切相关,可作为AML发病、复发及预后判断的有意义指标。

EXPRESSIONS OF MN1 AND PTEN GENES AND THEIR CLINICAL SIGNIFICANCE IN PATIENTS WITH ACUTE MYELOID LEUKEMIA

Abstract:Objective To explore the expressions of meningioma 1(MN1) and phosphatase and tensin homolog(PTEN) genes,and their relationship with the progression and prognosis of acute myeloid leukemia(AML). Methods RT-PCR technique was used to detect the expressions of MN1 and PTEN mRNA in mononuclear cells of 38 AML patients(16 cases in initial treatment group,12 in remission group and 10 in recurrence group).Thirteen bone marrow specimens from patients with nonmalignant hemopathy and normal individuals were served as controls. Results The positive expression rates of MN1 and PTEN gene in bone marrow mononuclear cells of AML patients were 76.3% and 60.5%,respectively.In initial treatment group,the expression level of MN1 gene was higher than that of the normal control group,while that of PTEN mRNA was significantly lower(F=2.385,2.054;q=2.305,2.867;P〈0.01);In remission group,the expression of MN1 mRNA level was significantly lower than that in initial treatment group,while PTEN mRNA level was significantly higher(q=2.121,2.756;P〈0.05);In the recurrence group,MN1 mRNA level increased and PTEN mRNA decreased again versus the normal control group(q=2.361,2.723;P〈0.05).The expressions of MN1 and PTEN were negatively correlated in AML(r=-0.321,P〈0.05). Conclusion The expressions of MN1 and PTEN genes are closely related to the occurrence and prognosis of AML,which can be used as significant parameters to predict the occurrence,recurrence and prognosis of this illness.
