抗炎清肾方三味单药对膜性肾病大鼠的保护作用

摘    要:目的探讨三味单味中药汤剂对膜性肾病大鼠的保护作用。方法将C-BSA诱发的32只存活的膜性肾病大鼠平分为模型组、百部组(百部治疗组)、白芷组(白芷治疗组)和白芨组(白芨治疗组),并以5只正常大鼠做空白对照。分别于实验第3、4、8周检测尿蛋白,于实验的第8周末采血后处死大鼠,检测血清肌酐、白蛋白、甘油三酯和胆固醇,观察肾组织病理改变,并用间接免疫荧光法检测各组大鼠肾组织内免疫球蛋白IgG、C3的表达水平。结果各中药组蛋白尿较模型组及治疗前均有改善(P<0.05)。百部和白芷组血清白蛋白比模型组水平增高(P<0.05):白芷组为(28.36±1.04)g/L,百部组为(29.83±1.98)g/L,模型组为(26.94±2.97)g/L。各中药组大鼠肾小球免疫复合物沉积较模型组有所减少、基底膜及系膜区的IgG、补体C3的沉积均较模型组减轻,尤以百部组和白芷组明显。结论中药抗炎清肾方中三味单味中药汤剂均对膜性肾病大鼠有一定治疗作用,百部和白芷的作用尤为明显。

Abstract:Objective To observe the protective effect of three kinds of Chinese herb in rats with membranous nephropathy(MN).Methods A glomerulonephritis model of immune complex in site was developed by cationic bovine serum albumin.The rats with MN were random assigned to model group and three Chinese herb groups,Stemona japonica group,Angelica dahurica group and Bletilla Striata group,healthy rats as normal control.Urine protein excretion was measured at the 3rd,4th,8th week.The rats were sacrificed at the end of the 8th week and serum creatinine,albumin,cholesterol and triglyceride were measured.Renal pathology changes were evaluated.Immunoglobulin G(IgG) and C3 in glomerulus were examine by immunofluorescence.Results Compared with model group,Stemona japonica group,Angelica dahurica group and Bletilla Striata group showed less amount of urinary protein excretion(P<0.05),Stemona japonica group and Angelica dahurica group showed higher serum albumin and less renal pathologic injury.Immunofluorescence staining indicated that there were less IgG and alexin C3 deposition in Stemona japonica group and Angelica dahurica group(P<0.05).Conclusion The results indicate that Stemona japonica,Angelica dahurica especially Stemona japonica have some protective effects in rats with MN.
