敌敌畏急性中毒小鼠脑ATP酶的活性检测

摘    要:目的 观察小鼠敌敌畏急性中毒后脑组织各区域的Ca^2+/Mg^2+-ATP酶和Na^+/K^+-ATP酶的活性水平,探讨敌敌畏中毒对中枢神经系统的影响。方法雄性小鼠16只.随机分成中毒组和对照组,中毒组按25mg/kg腹腔注射敌敌畏,对照组注射生理盐水。25min后处死,分别测定大脑、小脑和脑干组织内的Ca^2+/Mg^2+-ATP酶、N^a+/K^+-ATP酶的活性水平。结果脑组织中的ATP酶的活性水平都有不同程度的降低。在小脑,敌敌畏中毒组的两种ATP酶活性分别是对照组的62.11%。47.56%。在脑干,中毒组的两种ATP酶活性分别是对照组的67.72%,64.83%。在大脑的Ca^2+/Mg^2+-ATP酶和Na^+/K^+-ATP酶活性水平有下降。但两组相比无统计学差异性。结论敌敌畏中毒引起脑组织Ca^2+/Mg^2+-ATP酶和Na^+/K^+-ATP酶活性水平降低.共同导致脑细胞内钙稳态失衡。影响了神经递质的释放。

Abstract:Objective To study the activity of Ca~(2+)/Mg~(2+)-ATPase and Na~+/K~+-ATPase in acute dichlorvos-poisoned mice brain,and to understand the mechanism of its role in dichorvos poisoning.Methods Mice in poisoned group were administrated dichlorvos(25 mg/kg,ip),control group were administrated saline.After 25 min,The activity of Ca~(2+)/Mg~(2+)-ATPase and Na~+/K~+-ATPase were determined in cerebrum,cerebellum and brain stem.Results In acute dichlorvos-poisoned mice compared with the control group,the activity of Ca~(2+)/Mg~(2+)-ATPase decreased by 62.11% and(67.72%) in cerebellum and brain stem respectively,and Na~+/K~+-ATPase decreased by 47.56% and(64.83%.) The activity of Ca~(2+)/Mg~(2+)-ATPase and Na~+/K~+-ATPase was found to be declined,but there was no significance difference in cerebrum.Conclusion The results presented herein,indicate that the toxic effects of dichlorvos could be mediated through inhibiting the activity of Ca~(2+)/Mg~(2+)-ATPase and Na~+/K~+-ATPase,then through modifying the intracellular calcium homeostasis which may lead to affect the release of neurotransmitter.
