摘    要:目的 研究异烟肼对氯法齐明在小鼠体内组织分布和蓄积的影响.方法 70只昆明种小鼠采用随机数字表法分为氯法齐明单次给药组、异烟肼与氯法齐明联合单次给药组、氯法齐明连续给药组及异烟肼与氯法齐明联合连续给药组,单次给药组每组15只,连续给药组每组20只.给药剂量为氯法齐明13 mg/kg、异烟肼25 mg/kg,配成0.2 ml溶液灌胃.连续给药组每日给药1次,2个月后停药.单次给药组分别于给药后1、5和24 h,连续给药组分别于给药第30天和第60天以及停药第30天和第60天的次日,各时间点每次分别将每组5只小鼠及3只不给药小鼠摘眼球取血,解剖取其肝、脾、肺、肾、脂肪、小肠、皮肤等组织.组织匀浆处理,血浆样品乙腈沉淀蛋白后,应用高效液相色谱法测定氯法齐明的浓度.结果 单次给药时,氯法齐明单独应用及与异烟肼联合应用,小鼠肾、肺、脾和肝组织均在5 h达到峰值;脂肪组织中氯法齐明浓度持续增高,24 h最高,达(24.9±6.9)μg/g;血浆中药物浓度低于组织浓度.连续给药1个月后氯法齐明在组织和血浆中的浓度均明显高于单次给药,连续给药2个月后的氯法齐明浓度在脾、肾、肺等组织中明显高于连续给药1个月.连续给药1个月和2个月后脂肪中氯法齐明浓度均最高,在肾、脾、肝、肺、皮肤浓度亦较高(32~433μg/g),联合给药组肺脏中药物浓度明显高于氯法齐明组,1个月时分别为(32±8)和(57±11)μg/g,2个月时分别为(47±12)和(73±49)μg/g;脂肪组织中药物浓度明显低于氯法齐明组,1个月时分别为(433±53)和(289±30)μg/g,2个月时分别为(527±158)和(275±119)μg/g.两组停药后氯法齐明的浓度均逐渐降低,但停药2个月仍有一定的组织浓度.结论 氯法齐明在脂肪、脾、肾、肝、肺等组织分布比较高,异烟肼与氯法齐明合用可提高氯法齐明在肺组织中的浓度,减少脂肪中分布.

Abstract:Objective Tissue distribution and deposition of c]ofazimine in mice were investigated following administration of clofazimine with or without isoniazid. Methods Kunming mice were given clofazimine suspension orally at a daily dose of 13 mg/kg body weight either alone or with isoniazid (25 mg/ kg body weight) for a single dose or for 1 or 2 months. Tissues (liver, lung, spleen, small intestine, kidneys, fat) and pooled plasma were analyzed for clofazimine in all the treated groups by high-performance liquid chromatography. Results The levels of clofazimine in fat tissues, kidneys, spleens, livers, lungs and small intestine were the highest in mice receiving the drug continuously for 2 months, and were also higher in mice receiving the drug for 1 month as compared to mice receiving a single dose administration. After continuous administration for 1 or 2 months, the clofazimine level was the highest in fat tissues as compared to other tissue samples. The clofazimine level in the lungs was higher in mice receiving concomitant administration of isoniazid 1 month (57±11) μg/g, 2 months (73±49) μg/g] than in those receiving clofazimine alone 1 month (32±8) μ/g, 2 months (47±12) μg/g], but the clofazimine level in the fat tissue was significantly lower in mice receiving concomitant isoniazid 1 month (289±30) μg/g, 2 months (275±119) μ/g], than in those receiving clofazimine alone 1 month (433±53) μg/g, 2 months (527±158) μg/g]. Condusian Concomitant administration of isoniazid reduced clofazimine levels significantly in fat tissues while resulted in an increase of its level in lung tissues.
