HA-P软腭植入材料的研制及细胞毒性检测

摘    要:目的:研制一种新的治疗鼾症的软腭植入系统微创手术的生物材料并于体外检测其细胞毒性,探讨将其应用于软腭植入系统的可能性.方法:将羟基磷灰石颗粒(HA)、聚乳酸-三亚甲基碳酸酯P(LATMC)按一定比例复合,利用有机溶剂注模法,制备重组软腭植入材料;并采用MTT检测法,将不同浓度的HA-P浸提液与MH-3T3成纤维细胞接触1、3、5 d,检测细胞的增殖活性,计算细胞的相对增殖度,用6级毒性法进行评级.结果:研制不同浓度的HA-P材料浸提液在不同的时间点培养的细胞均正常增殖(P>0.05),毒性级别为0~1级.结论:研制的HA-P材料具有良好的细胞相容性,可能是一种良好的软腭植入系统材料.

Abstract:Objective:To explore a new material for palatal implant system and evaluate its cytotoxicity.Methods: organic solvent molding of hydroxyapatite and poly(lactide-co-trimethylene carbonate) proportionally mixed was performed to obtain palatal implant material.NIH-3T3 rat fibroblasts were exposed to different doses of HA-P extracts.After 1,3,5 days,cell proliferation and relative multiplication rate were calculated with MTT assay.Cytotoxicity was assessed by a 6-scale system.Results: cell proliferation was normal in the different extract at different time points.Levels of toxicity ranged from 0 to 1 grade.Conclusion: The HA-P material is compatible with tested cells,and therefore can be used for palatal implant system.
