胆红素对抗急性肺损伤大鼠的实验研究

摘    要:目的 探讨胆红素对急性肺损伤(ALI)大鼠的抗氧化指标和肺血管内皮细胞间黏附分子1(ICAM-1)表达的影响。方法 雄性Wistar大鼠30只,随机分为对照组、ALI模型组、胆红素干预组。检测肺组织超氧化物歧化酶(SOD)、脂质过氧化产物丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH—Px)含量变化;测定肺血管内皮ICAM-1 mRNA的表达。结果 ALI模型组MDA和ICAM-1 mRNA显著高于对照组(分别为P〈0.01和P〈0.001),而SOD和GSH-Px明显低于对照组(均为P〈0.01)。胆红素干预组MDA和ICAM-1 mRNA明显低于ALI模型组(均为P〈0.01),而SOD和GSH-Px显著高于ALI模型组(分别为P〈0.01和P〈0.05)。结论 胆红素通过抗氧化作用和抑制ICAM-1 mRNA的表达对抗ALI。

Abstract:Objective To investigate the effect of bilirubin on acute lung injury (ALI) of oxidative/antioxidative parameters and ICAM-1 mRNA expression in pulmonary vascular endothelial cells. Methods Thirty male Wistar rats were randomly divided into three groups:control group, ALI model group and pre-treatment bilirubin group. Oxidative/antioxidative parameters and ICAM-1 mRNA expression were determined. Results In ALI model group,MDA content and ICAM-1 mRNA expression in pulmonary vascular endothelial cells were significantly higher than those in control group(P<0.01 and P<0.001), but the level of SOD and GSH-Px were significantly lower than those in control group (P<0.01). In pre-treatment bilirubin group, MDA content and ICAM-1 mRNA expression were significantly lower than those in ALI model group(P<0.01,P<0.001),but the activities of SOD and the content of GSH-Px were significantly higher than those in ALI model group(P<0.01,P<0.05). Conclusion Bilirubin might significantly alleviate the degree of ALI by antioxidation and inhibit excess production of ICAM-1 mRNA.
