摘    要:目的 探讨A型肉毒杆菌毒素 (botulinumtoxintypeA ,BTA)对豚鼠鼻腔黏膜腺细胞凋亡的影响。方法  1 8只雄性豚鼠随机分为BTA组和对照组。BTA组左侧鼻腔以浸有 1 0U (0 .2ml)BTA的Merocel作用 1h ,对照组以生理盐水代替BTA。分别于用药后第 1周、2周、4周全麻下切取豚鼠左侧下鼻甲黏膜 (每组 3只 ) ,4 %多聚甲醛固定后石蜡包埋。切片行末端脱氧核苷酸转移酶介导的dUTP缺口末端标记 (TUNEL)和AB PAS染色。结果 BTA局部应用第 1、2周豚鼠鼻黏膜腺体及腺上皮中凋亡细胞显著增加 (P <0 .0 1 ) ,第 4周凋亡指数恢复正常。细胞凋亡主要发生在浆液腺。结论 BTA可诱导豚鼠鼻黏膜浆液腺细胞凋亡。

Abstract:Objective To investigate the effect of botulinum toxin type A (BTA) on apoptosis in nasal glands of guinea pig. Methods Eighteen male guinea pigs were randomly divided into BTA group and a control group. Merocel sponge soaked with 10U ( 0.2 ml) BTA was put into left nasal cavity of guinea pig under general anesthesia for one hour in BTA group. Saline replaced BTA in the control group. The inferior turbinate mucosae of guinea pig were harvested under general anesthesia at 1, 2, 4 weeks after BTA treatment (3 animals in each group). The tissues were fixed in 4% paraformaldehyde and paraffin embedded. Apoptosis was detected with terminal deoxyuncleotidyl transferase-mediated deoxyuridine triphosphate nick end labeling (TUNEL) staining and the types of gland cells were determined with AB-PAS staining. Results One and two weeks after local BTA application, apoptosis of glandular and canal cells in nasal mucosa of guinea pigs increased significantly (P< 0.01). The apoptosis index became normal at 4 weeks after BTA treatment. Apoptosis mainly occurred in serous gland. Conclusion BTA can induce apoptosis in nasal serous glands of guinea pig.
