慢性间歇性低氧对大鼠胸主动脉内皮细胞的影响

摘    要:目的 探讨慢性间歇性低氧对大鼠胸主动脉内皮细胞的损伤变化。方法 将12只10~12周龄的雄性SD大鼠采用抽签法分为对照组和实验组,每组各6只。实验组大鼠每天10点至18点置于间歇性低氧环境中暴露,对照组大鼠不做低氧处理,共4周。饲养时间结束后取大鼠胸主动脉标本行苏木精-伊红染色(hematoxylin and eosin staining,HE染色)及原位末端转移酶标记法(terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling assay,TUNEL)染色观察细胞病变情况,并计算病变细胞、凋亡细胞在视野中所占比例。结果 HE染色光镜下实验组可见血管内皮细胞肥大,排列不规则,细胞核、胞质着色淡,平滑肌细胞核肿胀;对照组见血管内皮细胞和平滑肌细胞排列规则、细胞核形态及染色正常。TUNEL染色光镜下实验组可见多个凋亡细胞,对照组偶见凋亡细胞。实验组大鼠主动脉组织的细胞病变率及凋亡率均显著高于对照组(P<0.01)。结论 阻塞性睡眠呼吸暂停低通气综合征所产生的慢性间歇性低氧可损伤大鼠胸主动脉...

Effects of chronic intermittent hypoxia on endothelial cells of thoracic aorta in rats
Abstract:Objective To investigate the injury changes of thoracic aorta endothelial cells induced by chronic intermittent hypoxia in rats. Methods Twelve male SD rats aged 10 to 12 weeks were divided into control group and experimental group by drawing lots, with 6 rats in each group. The rats in experimental group were exposed to intermittent hypoxia from 10:00 to 18:00 every day for 4 weeks, and rats in control group were not treated with hypoxia. At the end of feeding time, the thoracic aorta samples of rats were taken for hematoxylin and eosin staining (HE staining) and terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling assay (TUNEL) staining to observe the cytopathic effect. Results HE staining showed hypertrophy of vascular endothelial cells, irregular arrangement, light staining of nucleus and cytoplasm, and swelling of smooth muscle cells. In control group, vascular endothelial cells and smooth muscle cells were arranged regularly, their nuclear morphology and staining were normal. Many apoptotic cells were observed in experimental group and occasionally in control group under TUNEL staining. The cytopathic rate and apoptosis rate of aorta in experimental group were significantly higher than those in control group (P<0.01). Conclusion Chronic intermittent hypoxia induced by obstructive sleep apnea hypopnea syndrome can damage vascular smooth muscle cells and induce apoptosis of vascular endothelial cells in the thoracic aorta of rats.
