摘    要:目的:研究实验性脊髓损伤后低氧诱导因子(HIF-1α)的时序性变化及与创伤性凋亡的关系,探讨其在脊髓损伤中的意义。方法:以大鼠静压型脊髓损伤模型为研究对象,运用免疫组化方法进行HIF-α染色,流式细胞术测定凋亡率及bcl-2的表达。结果:从脊髓损伤后1天开始,HIF-1α表达明显增加,3-7天达到高峰,14天时HIF-1α阳性染色率下降显著,HIF-1α的时序性变化与凋亡率和bcl-2含量的变动趋势密切相关。结论:HIF-1α可能参与了脊髓损伤后缺血缺氧的继发损害过程,它与创伤后凋亡之间有着密切的关系,对其进行深入研究可能为找到有效的治疗脊髓损伤的方法提供新的思路。

Abstract:OBJECTIVE: To investigate the sequential changes of hypoxia-inducible factor 1 alpha (HIF-1 alpha) in experimental spinal cord(SCI) injury in rats and analyze its potential effects on SCI. METHODS: Static compression model of SCI was employed in this study. Expressions of HIF-1 alpha were measured with immunohistochemical staining. Flow cytometry was used to determine the apoptotic ratio and bcl-2 expressions. RESULTS: HIF-1 alpha began to increase one day after injury, and reached peak at 3-7 days. Two weeks later, it declined significantly. The sequential changes of HIF-1 alpha coincided well with the alterations of apoptotic ratio and bcl-2 content. CONCLUSION: HIF-1 alpha may participate in the secondary ischemic and hypoxic procedures after spinal cord injury and mediate the traumatic apoptosis. Further understanding of HIF-1 alpha may provide new therapeutic regimens for SCI.
