脉冲电磁场对大鼠骨髓间充质干细胞体外诱导分化影响的研究

摘    要:目的观察脉冲电磁场(PEMFs)对5-氮胞苷(5-aza)诱导大鼠骨髓间充质干细胞(MSCs)向心肌样细胞分化的影响。方法50Hz脉冲低频电磁场刺激5-aza诱导7d后的MSCs,根据不同的感应强度和作用的时间分为A、B、C组和1、2、3、4亚组,设未暴露PEMFs的为对照组。通过Western印迹法检测不同条件下肌钙蛋白T(cTnT)的表达量的变化。结果在0.5、1、5mT组中20、30min各亚组较对照组有显著性增加,在5mT组在10min的cTnT的表达量显著性减少。结论50Hz的0.5~5mT感应强度的PEMFs作用20~30min为促进体外5-aza诱导MSCs向心肌方向分化的有效窗口。

Abstract:Objective To investigate the effects of pulsed electromagnetic fields on the differentiation of rat marrow mesenchymal stem cells induced with 5-azacytidine.Methods Rat marrow mesenchymal stem cells induced for 7 days with 5-azacytidine was exposed or unexposed to 50Hz pulsed electromagnetic fields,and then randomly was divided into A,B,C three group and 1,2,3,4 four subgroup accroding to different inductance intensity and different exposed time,Unexposed under pulsed electromagnetic fields was control groups.The protein expression of cTnT was determined by Western blotting.Results The expressions of cardiac troponin T enhanced significantly under 0.5,1mT and 5mT inductance intensity for 20-30min and reduced significantly under 5mT inductance intensity for 10min by Weston blot.Conlunsion Under 0.5mT and 1mT inductance intensity for 20-30min of 50Hz pulsed electromagnetic is effective condition that can promote marrow mesenchymal stem cells differentiate into cardiomyocyte-like cells.
