摘    要:目的:观察PKA-CREB信号转导通路在慢性脑缺血所致大鼠认知功能损害中的作用。方法:结扎大鼠双侧颈总动脉,制成慢性脑缺血模型,分缺血8周组和假手术对照组,术后第8周时用Morris水迷宫测定大鼠学习记忆能力。用Westernbloting检测大鼠海马胞核内PKAca及pCREB的表达。结果:缺血组与假手术组相比学习记忆能力明显下降(P<0.05),缺血组大鼠海马中PKAca及pCREB的表达与假手术组相比也下降(P<0.05),且两者之间的下降存在正相关关系。结论:PKA-CREB信号转导通路可能参与了慢性脑缺血所致大鼠认知功能的损害。

Abstract:Aim: To study the effects of PKA-CREB signal transduction system on cognitive impairment in rats with chronic cerebral hypoperfusion. Methods: The chronic cerebral hypoperfusion model was made by permanent bilateral occlusion of both common carotid arteries, and the rats were randomly divided into sham control and ischemic groups. The spatial learning and memory function was tested with Morris water maze at the eighth week after the operation. The levels of PKAca and pCREB in hippocampus tissues of rats were measured by Western blotting. Results: The learning and memory ability in rats was significantly injured in the ischemic group. The levels of PKAca and pCREB in the ischemic group were lower than that in the sham control group. The protein levels PKAca and pCREB were positively correlated with learing and memory performance in Morris water maze test. Conclusion: The PKA-CREB signal transduction system may be involved in the spatial learning and memory function in the rats with chronic cerebral hypoperfusion.
