穿心莲指纹图谱和不同产地质量研究

摘    要:目的:建立穿心莲药材有效的质量评价方法.方法:通过建立HPLC指纹图谱和有效成分的含量测定,对来自10个不同产地的穿心莲药材进行分析.色谱条件为Alltima C18色谱柱(4.6m×250mm, 5μm),流动相为乙腈-水系统,梯度洗脱,流速1.0mL·min-1,指纹图谱检测波长为215nm,穿心莲内酯和脱水穿心莲内酯含量测定的检测波长分别为225nm和254nm.结果:得到分离度、重现性均较好的穿心莲药材HPLC指纹图谱,标示了10个共有色谱峰,对10批来自不同产地穿心莲样品的色谱指纹图谱进行了相似度评价;应用HPLC-DAD测定了不同产地穿心莲样品中穿心莲内酯和脱水穿心莲内酯的含量,结果显示,穿心莲药材中穿心莲内酯和脱水穿心莲内酯总量介于1.09%~4.47%.结论:该方法简便准确,灵敏度高、重复性好,可以作为穿心莲药材的质量控制方法,此方法已被<香港中药材标准>采用.建议HPLC指纹图谱用于穿心莲药材的质量控制,穿心莲内酯和脱水穿心莲内酯的总量不得低于1.0%.
