摘    要:目的]评价银杏叶提取物(EGb761)对神经胶质瘤U87细胞的生长与侵袭的影响,并进一步分析EGb761对U87细胞作用的分子机制。方法]体外培养人神经胶质瘤U87细胞,给予不同浓度(0、100、200、400mg/L)银杏叶提取物干预(24h、48h、72h)后,采用CCK-8技术检测EGb761对U87细胞增殖的影响。Transwell小室和流式细胞凋亡技术分析EGb761对U87细胞侵袭和凋亡的影响;Western blot检测EGb761处理后U87细胞内NF-κB与cyclin D1、 iNOS和COX-2蛋白的表达变化。结果]银杏叶提取物可有效抑制神经胶质瘤U87细胞的增殖和侵袭,促进细胞凋亡;Western blot提示EGb761干预的U87细胞内NF-κB与cyclin D1、 iNOS、COX-2蛋白的表达均明显下调。结论] EGb761抑制神经胶质瘤U87细胞的生长增殖和侵袭能力,其可能是通过NF-κB信号通路而发挥抗肿瘤作用的。

Abstract:Objective] To investigate the effect of Ginkgo biloba extract(EGb761) on the growth and invasion of glioma U87 cells and the molecular mechanism. Methods] U87 cells were cultured in vitro and treated with EGb761 of different concentrations(0,100,200,400 mg/L) for different time(24 h,48 h,72 h). The proliferation of U87 cells was detected with CCK-8 assay. Transwell migration assay and flow cytometry were employed to determine the effect of EGb761 on invasion and apoptosis of U87 cells,respectively. Changes of NF-κB,cyclin D1,i NOS and COX-2 expressions in U87 cells were detected by Western Blot. Results] EGb761 inhibited the proliferation and invasion of U87 cells and promoted its apoptosis. Western blot revealed that EGb761-treated U87 cells significantly down-regulated the expression of NF-κB,cyclin D1,i NOS and COX-2.Conclusion] EGb761 can significantly inhibit the proliferation and invasion of U87 cells,which is associated with the regulation of NF-κB signaling pathway.
