接骨宝对实验性骨折愈合过程中BMP-2表达的影响

摘    要:目的:通过观察骨形态发生蛋白-2(BMP-2)在骨折愈合过程中的表达及分布情况,探讨接骨宝对实验性骨折愈合过程中BMP-2表达的影响和促进骨折愈合的机制。方法:制作40只兔桡骨骨折模型.随机分为接骨宝组及对照组,术后1d接骨宝组即喂服接骨宝,对照组喂服白开水。分别于术后1、2、3、4、5周取样,随后进行组织学切片及BMP-2免疫组化染色,并采用图像分析系统对BMP-2的表达进行分析。结果:①在骨折愈合过程中。两组在骨痂处的BMP-2含量的总体变化趋势相同,均为早期高后期低,用药组的BMP-2高峰值出现在第2周.对照组出现在第3周;②接骨宝组在整个愈合过程中BMP-2的含量均高于同期的对照组。结论:接骨宝能影响骨折愈合过程中BMP-2的合成、分泌、分布和衰减,进而调节成骨细胞、软骨细胞的分化,促进骨细胞的成熟,增强破骨细胞的活性,加速骨折的修复和塑形,缩短了骨折愈合的进程。

Influence of Jiegubao on the expression of BMP-2 during the process of experimental fracture healing

Abstract:Objective: To investigate the influence of Jiegubao on the expression of bone morphogenetic protein-2 (BMP-2) and mechanism of fracture healing promoted by Jiegubao by observing the expression and distribution of BMP-2 during fracture healing. Methods: The 40 model rabbits of fracture of radius were prepared and randomly divided into Jiegubao group and control group. The model rabbits of Jiegubao group were treated with decoction of Jiegubao by intragastric administration, and the rabbits of control group were treated with plain boiled water. The samples of all the model rabbits were collected respectively 1, 2, 3, 4 and 5 weeks after the operation of fracture of radius respectively. The samples of radius were observed by the method of histology, and BMP-2 was detected by immunohistochemistry. Expression of BMP-2 was analyzed by image analysis system. Results: (1)There were identical total change tendency of BMP-2 contents of two group osteotylus during fracture healing. BMP-2 contents of osteotylus were plentiful in earlier period, and were low in later period. High-peak of BMP-2 contents of Jiegubao group occurred in the second week, and that of control group occurred in the third week. (2)BMP-2 contents of Jiegubao group were surpassed to that of control group during the whole process of fracture healing. Conclusion: Jiegubao can adjust composition, secretion, distribution and decrescence of BMP-2 during fracture healing, regulate cell differentiation of osteoblast and chondrocyte, promote the mature of bone cell, strengthen the activity of osteoclast, accelerate renovation and moulding of fracture and shorten the proceeding of fracture healing.
