摘    要:以S.lincolnensis B48为对照,分别测定两个染色体上含lmbI基因灭活拷贝的菌株YYl(同源交换)、YY2(同源整合)的抗菌效价,其中YYl在其整个生物周期中均无明显抗菌效价,而YY2的效价与B48相比也明显伯低。进一步通过PCR反应验证这两株基因工程菌lmbI基因灭活情形,结果发现YYl染色体上lmbI基因的两个拷贝均因同源交换而被灭活,而YY2可能是因单拷贝发生同源整合而仅被部分灭活。

Identification of Homologous Recombination of Lincomycin Biosynthesis Defective Strains

Abstract:Strain YY1 and YY2 were two genetic recombinant strains derived from S.lincolnensis B48 by homologous exchange and homologous integration, respectively. Compared with B48, the potency of lincomycin of YY2 was much lower, while that of YY1 turned to be zero. The genetic recombination was further identified by PCR and the result showed that two copies of lmbI genes in YY1 were inactivated by homologous exchange, while probably only one copy in YY2 was inactivated by homologous integration and the other remained intact.
