针刺对大鼠脑缺血再灌注损伤后ICAM-1表达的影响

摘    要:目的 观察针刺对大鼠局灶性脑缺血再灌注损伤后脑组织中细胞间粘附分子-1(intercellular adhesion molecule-1,ICAM-1)表达的影响,探讨针刺在脑缺血再灌注损伤中的保护作用机制。方法 采用线栓法制备大鼠大脑中动脉栓塞(MCAO)模型,应用免疫组化方法观察假手术组、模型组(再灌注6h、24h、72h)、针刺组(再灌注6h、24h、72h)ICAM-1表达的变化。结果 缺血再灌注后6h,脑组织微血管内皮细胞表达ICAM-1出现增高趋势,并于24h达到高峰,模型组与针刺组及模型组与假手术组间均有显著差异(P〈0.01或P〈0.05)。结论 脑缺血后1CAM—l表达增加,针刺可降低1CAM.1的表达,从而减轻缺血后ICAM—l导致的缺缺血性脑损伤。说明针刺对缺血性腑损伤的保护作用机制可能与其抑制脑内ICAM-1的表达有关。

Abstract:Objective To observe the influence of acupuncture on cerebral expression of intercellular adhesion molecule-1(reperfusion injury) and investigate the mechanism of its protective action in rats with cerebral ischemia and reperfusion injury. Methods A rat model of middle cerebral artery occlusion(MCAO) was made by thread occlusion.A change in ICAM-1 expression was observed by immunohistochemistry in sham operation,model(6,24 and 72 h after reperfusion) and acupuncture(6,24 and 72 h after reperfusion) groups.Results ICAM-1 expression in cerebral microvascular endothelial cells tended to increase at 6 h after reperfusion and peaked at 24 h.There were significant differences between model and acupuncture groups and between model and sham operation groups(P<0.01 or P<0.05).Conclusion ICAM-1 expression increases after cerebral ischemia.Acupuncture can decrease ICAM-1 expression and thereby reduce ischemic brain injury due to post-ischemic ICAM-1.It is indicated that the mechanism of the preventive action of acupuncture on ischemic brain injury may be related to its inhibition of cerebral ICAM1 expression.
