摘    要:该研究应用培养人脐静脉内皮细胞株ECV304在5,20,及5/20mmol/L葡萄糖浓度条件下,通过MTT,AnnxenV-FITC检测细胞生存抑制率及凋亡率观察,探讨葡萄糖漂移对血管内皮细胞的作用。结果显示恒定性高葡萄糖细胞生存率低于正常浓度,而波动葡萄糖浓度的生存率较恒定高浓度葡萄糖更低。结论:血糖漂移对血管内皮细胞的损害较恒定性高血糖更严重。是独立于恒定性高血糖之外的血管内皮细胞伤害因素。

Abstract:Objective To explore the effect of fluctuating glucose on ECV304.Method Its cells were incubated for 14 days in media containing different concentrations;5mmol/L,20mmol/L,or adaily alternating 5 or 20 mmol/L glucose.Apoptosis was studied by different methods;viability assay,MTT colorimetric assay,acridine or-ange and flow cytometry assay.Results Intermittent high glucose induces ECV304 apoptosis,furthermore the levels of apoptosis cells enhance more than stable high glucose.Conclusion Variability in glycemic control could be more deleterious to endothelial cells than a constant high concentration of glucose.
