缺血预处理对缺血再灌注后兔腰髓组织中HSPs-70 mRNA表达的影响

摘    要:为研究缺血预处理对缺血再灌注后兔腰髓组织中HSPs-70mRNA表达的影响,探讨缺血预处理脊髓缺血再灌注损伤保护作用的机制,经股动脉向腹主动脉内置入4F Swan-Ganz导管,导管气囊置于左肾动脉开口远端,0.5-1.0cm,向气囊内注气建立脊髓缺血模型。将实验兔分为假手术组,对照组和预处理组,应用RT-PCR方法对3组兔缺血再灌注后不同时间点脊髓组织中HSPs-70mRNA表达进行观察。结果发现,假手术组兔的脊髓组织没有HSPs-70mRNA的阳性表达,缺血再灌注后,预处理组兔脊髓组织的HSPs-70mRNA表达较对照组明显增强(P<0.01),表达时间也显著延长。提示缺血预处理可以明显增强缺血后脊髓组织中HSPs-70mRNA的表达,延长HSPs-70mRNA的表达时间,并可能通过此机制对缺血脊髓产生保护作用。

Abstract:To study the effect of ischemic preconditioning(IP) on the expression of HSP 70 mRNA in postischemic spinal cord of rabbits, the spinal cord ischemic model was established by inflating the balloon of a 4F Swan Ganz catheter positioned in the abdominal aorta of the rabbits. The rabbits were divided into sham operated group, ischemic group and IP group. The expression of HSP 70 mRNA in postischemic spinal cord was observed by using the method of RT PCR at 4 hours, 2 days and 5 days after spinal cord ischemia respectively. No expression of HSPs 70 mRNA was found in sham operative group. Compared with ischemic group,the expression of HSP 70 mRNA in spinal cord of IP group was strengthened( P <0 01) and its expression time was prolonged. It is suggested that ischemic precontioning can enhance the expression of HSPs 70 mRNA postischemic spinal cord of rabbits and prolong the expression time of HSPs 70 mRNA. It may protect spinal cord from ischemia through this mechanism.
