COX-2抑制剂塞来昔布对大鼠脊髓损伤的保护作用

摘    要:目的 观察选择性环氧化酶一2抑制剂塞来昔布(celecoxib)对大鼠脊髓损伤的保护作用。方法 48只SD大鼠,随机分为单纯损伤组及治疗组。采用改良Allen’s打击法制作脊髓损伤动物模型,分别在伤后12、24、72h测定脊髓标本的丙二醛(MDA)和超氧化物歧化酶(SOD)含量。用苏木精-伊红染色观察损伤脊髓组织病理变化,原位末端标记法(TUNEL)标记凋亡细胞。同时采用Tarlov评分和斜板实验观察脊髓神经功能恢复情况,比较各组之间的差异。结果 治疗组较单纯损伤组神经功能明显改善(P〈0.05)。治疗组各时间点MDA含量较单纯损伤组明显降低(P〈0.05),SOD含量明显升高(P〈0.05),病理形态改变较轻且凋亡细胞少(P〈0.05)。结论 COX-2抑制剂塞来昔布可减少神经细胞凋亡,对继发性脊髓损伤起到保护作用。

Abstract:Objective To investigate the protective effect of celecoxib on secondary spinal cord injury(SCI).Methods Forty-eight rats were divided randomly into two groups:injury group and treatment group.By using modified Allen's contusion method,spinal cord injury model was established.At 12th,24th and 72nd h after injury,MDA and SOD contents in spinal specimens were determined.The pathological changes in the injured spinal cord tissue were observed by hematoxylin and eosin(HE) staining.The apoptotic cells were labeled with terminal doxynucleotidyl transferase-mediated dUTP nick end labeling(TUNEL) method.The inclined plane and Tarlov scores were used to assess the recovery of spinal cord neurological function.Results Behavioral score in treatment group was significantly higher than that in injury group(P<0.05).As compared with injury group,MDA was decreased,SOD increased significantly(P<0.05),pathological changes alleviated and the apoptotic cells reduced in treatment group at different time points(all P<0.05).Conclusion Celecoxib had protective effects on the secondary spinal cord injury in rats by reducing apoptosis of neural cells.
