蜂胶超临界CO_2萃取物对模型大鼠胃溃疡的预防作用及其机制

摘    要:目的研究蜂胶超临界CO2萃取物(SCEP)对实验性大鼠胃溃疡的预防作用并探索其作用机制。方法采用水应激和乙醇诱导制备大鼠胃溃疡模型,观察灌胃SCEP及对照(植物油)、阳性对照(奥美拉唑)、蜂胶乙醇提取物对胃溃疡的影响,通过测定胃溃疡指数和治疗比例评价预防效果,通过测定大鼠血液中MDA含量及SOD活性探讨其抗胃溃疡的作用机制。结果 SCEP对两种模型大鼠的胃溃疡有明显的预防作用,效果明显优于蜂胶乙醇提取物、接近药物奥美拉唑,植物油对胃溃疡有一定的预防效果;蜂胶超临界CO2萃取物预防两种模型大鼠的最佳剂量分别为10 mg/kg和3 mg/kg;按照最佳剂量灌喂,大鼠血液中MDA含量分别比空白组降低38.30%和51.72%、SOD活力分别比空白组增加55.74%和71.89%。结论蜂胶乙醇提取物与SCEP均能提高水应激及乙醇诱导的急性胃溃疡大鼠血清和胃组织中SOD的含量,降低MDA水平,从而显示其有效降低氧自由基及愈合大鼠胃粘膜的能力。

Abstract:Objective To study the preventive effect of supercritical CO2 extract of propolis(SCEP) on gastric ulcer in rats and its mechanism.Method Water-stress-induced and ethanol-induced gastric ulcers were developed in rats.Then the rats were divided into control(plant oil),positive(Omeprazole) and ethanol extract of propolis groups.The MDA content and SOD activity in blood were determined.Results SCEP had obvious preventive effect on two kinds of gastric ulcers,and obviously better than propolis ethanol extract(P<0.05) and close to Omeprazole group(P<0.05).Plant oil had certain preventive effect on gastric ulcer.Optimal dosages of SCEP for prevention of two kinds of gastric ulcers were 10 mg/kg and 3 mg/kg.Compared with the control,blood MDA content in rats with two kinds of gastric ulcers decreased 38.30% and 51.72% respectively,and the SOD activity increased 55.74% and 71.89% respectively.Conclusion Both alcohol extact and SCEP have the effect of increasing SOD activity and decreasing MDA level in blood and gastric tissues in water-stress-induced and ethanol-induced gastric ulcer rat models,and show their effect in scavenging oxygen radicals and gastric mucosa recovery.
