RP-HPLC测定哈蟆油中1-甲基海因的含量

 目的建立哈蟆油中1-甲基海因反相高效液相色谱(RP-HPLC)含量测定方法。方法采用ZORBAX Eclipse XDB-C₁₈柱(4.6 mm×150 mm,5μm)色谱柱,检测波长为235 nm,乙腈-水(10∶90)为流动相,流速为0.3 mL·min^-1。结果在0.32～1.58μg内峰面积与其浓度呈良好的线性关系(r=0.999 9),方法的精密度RSD为0.8%。平均回收率为99.5%,RSD为1.3%。结论测定方法简便快捷、精密度高、准确性好,可以作为哈蟆油中1-甲基海因的测定方法,以控制其原料的质量。

Study on Determination of 1-Methyl Hydantoin in Oviductus ranae by RP-HPLC Assay

OBJECTIVE To establish a RP-HPLC method for the determination of 1-methyl hydantoin in Oviductus ranae.METHODS The determination was carried out with ZORBAX Eclipse XDB-C₁₈(4.6 mm×150 mm,5 μm),detected at the wavelength of 235 nm,and the mobile phase consisted of acetonitrile-water(10∶90) at a flow rate of 0.3 mL·min^-1.RESULTS The calibration curve of 1-methyl hydantoin was in good linearity over the range of 0.32～1.58 μg(r=0.999 9).The RSD value of precision was 0.8%.The average method recovery was 99.5% and RSD was 1.3%.CONCLUSION The method is simple,sensitive,accurate and reproducible.It can be used for determining the content of 1-methyl hydantoin in Oviductus ranae and can be used to control the qualities of Oviductus ranae.
