纳洛酮抗颅脑损伤后细胞凋亡及信号传递机制的实验研究

摘    要:目的 观察纳洛酮对严重颅脑损伤后神经细胞凋亡的影响,探讨Caspase -3介导的信号传递机制。方法 选用健康普通家兔随机分成正常组、对照组和纳洛酮组,分别予非致伤、致伤、致伤后纳洛酮腹腔注射处理,应用免疫组织化学方法检测各组家兔致伤部位和对侧非致伤部位脑组织中凋亡及Caspase -3蛋白表达阳性的神经细胞数量。结果 在致伤部位或对侧非致伤部位,正常组、纳洛酮组及对照组脑组织中调亡的及Caspase -3蛋白表达阳性的神经细胞所占比例的差异有统计学意义(P〈0.05,P〈0.01),数值大小为:正常组〈纳洛酮组〈对照组。结论 纳洛酮可抑制严重颅脑损伤后神经细胞凋亡,Caspase -3介导的信号传导机制可能参与纳洛酮抗严重颅脑损伤后神经细胞凋亡的过程。

Abstract:Objective To investigate the anti-apoptotic effect of naloxone in experimental severe traumatic brain tissue and to discuss Caspase-3-mediating-mechanism.Methods The healthy rabbits were divided randomly into normal group,control group and naloxone group.Rabbits of normal group did not been injured,control group were injured,and naloxone group got naloxone injection intraperitoneally after being injuried,respectively.Apoptosis and Caspase-3were detected by immunohistochemisty.The positive cells of TUNEL and Caspase-3were counted under light microscope and then the labeling index(LI)were calculated.Results The differences of the number of apoptotic nervous cells and Cas-pase-3-positive-nervous cells among each group were found very clearly(P<0.05,P<0.01),values were aligned from the least to the most:normal group,naloxone group and control group.Conclusion Naloxone can suppress apoptosis in severe traumatic nervous cells,and mechanismwith involvement of caspase-3may participate in the anti-apoptotic effect of naloxone in experimental severe traumatic brain tissue.
