PCR结合银染技术分析pMCT118多态性

目的　了解广西汉族、壮族人群pMCT118遗传多态性。方法　应用PCR结合聚丙烯酰胺凝胶电泳及银染技术对150名汉族无关个体及300名壮族无关个体进行基因扩增分型。结果　汉族人群中检出21个等位基因、63个基因型，壮族人群中校出23个等位基因、74个基因型。汉族人群杂合度、非父排除率、个体识别力及多态信息含量分别为0．873、0．770、0．973、0．879，而壮族则分别为0．824、0．762、0．968、0．875。广西汉、壮族人群pMCT118基因频率分布的差异无显性(P＞0．1)，且两个群体基因型频率分布均符合Hardy—Weinber8平衡。结论研究显示pMCT118位点为高度多态的遗传标记，在个体识别、亲权鉴定、群体遗传及器官移植等方面均有重要的应用价值。

Polymorphic analysis of pMCT118 locus by PCR

Objective To learn the distribution of allele and genotype frequencies of pMCT118 VNTR locus in Han and Zhuang ethnic groups in Guangxi. Methods 150 unrelated individuals of Han and 300 unrelated individuals of Zhuang were analyzed by PCR and PAGE followed by silver stain. Results 21 alleles and 63 genotypes were observed in Han population, 23 alleles and 74 genotypes were observed in Zhuang population. The observed heterozygosity (H), probability of exclusion (P E),discrimination power (DP) and polymorphism information content (PIC) were 0 873,0 770,0 973,0 879 in Han population and 0 824,0 762,0 968,0 875 in Zhuang population. No significant difference in the distribution of allele frequencies was observed between the two populations ( P >0 1). The observed genotype distribution of both population was in accordance with Hardy Weinberg equilibrium. Conclusion The results demonstrated that pMCT118 locus is a highly polymorphic marker for individual identification, paternity testing, genetic study of population as well as anthropology. [
