急性自体血栓肺栓塞大鼠动物模型制备

摘    要:目的:用自体血栓模拟自然发生的静脉血栓注入肺动脉,制备急性肺栓塞大鼠动物模型.方法:采用改良右心导管法将大鼠自体血栓注入肺动脉导致急性肺栓塞的发生.通过控制注入血栓的数量、速度,同时动态监测肺动脉压、体动脉压变化.用肺灌注扫描显像(emission computed tomography,ECT)和苏木素-伊红(hematoxylin-eosin,HE)染色组织光镜来观察判断造模成功与否.以肺系数法测肺水肿程度.结果:PE组0.5~4 h大鼠肺动脉压显著升高,体动脉压显著降低,PE组与C组比较差异有显著性(P<0.01);PE组2~4 h和C组比较,肺系数显著升高(P<0.01);ECT显像见肺组织明显呈放射性充盈缺损和稀疏,HE染色光镜下见注入血栓团块.结论:本模型各项变化符合急性肺栓塞的病理生理基础变化,是一有效、稳定的肺栓塞动物模型.

Abstract:Objective: To establish the rat model of acute autoblood pulmonary embolism when thrombus was injected into pulmonary artery with simulated natural vein thrombus. Methods: Thrombis of autoblood were injected to pulmonary artery through the modified right heart catheterization to induce acute autoblood pulmonary thromboembolism (PTE). By controlling quantity of thrombus and speed of injection, the changes of mean pulmonary arterial pressure(mPAP) and mean carotid arterial pressure (mCAP) of rat were dynamically monitored to judge the degree of the pulmonary thromboembolism. The established model was evaluated by emission computed tomography(ECT) and hematoxylin-eosin(HE) staining. Results: The mPAP was increased significantly after infusion of autoblood thrombs in PE group compared with C group(P<0.01); The mCAP was decreased obviously in PE group compared with C group(P<0.01).There was a significant increase in the level of coefficient of lung. Filling defect was showed significantly in ECT and the infused thrombi were witnessed by hematoxylin and eosin staining. Conclusion: The pathological and physiological changs of experimental rat model accord with the acute pulmonary thromboembolism's. It is a useful and steady mode
