摘    要:目的探讨VitC与三氧化二砷(As2O3)促喉癌细胞株Hep-2凋亡的协同效应。方法体外培养人喉癌细胞株Hep-2,以As2O3合用VitC孵育细胞,采用四甲基偶氮唑(MTT)法,Annexin-V/PI双染色流式细胞术来观察各组的细胞凋亡情况,并通过流式软件分析细胞周期变化。结果As2O3与VitC联用能显著降低单用As2O3的细胞存活率和升高细胞凋亡率(P<0.05),VitC能显著增强As2O3诱导细胞集中于G2/M期的作用,从而增强其促细胞凋亡作用。结论联用VitC能增强As2O3诱导细胞凋亡的作用,增强作用可能与影响细胞周期有关。

Abstract:Objective To study synergic effect of arsentic trioxide(As2O3) with vitamin C(VitC)on human laryngocarcinoma Hep-2 cell line apoptosis.Methods Human laryngocarcinoma cell line Hep-2 was cultured in vitro,and incubated with As2O3or jointly with As2O3 and VitC;the inhibition abiliy of Hep-2 cells was determined by MTT,the apoptosis was mensurated by the flow cytometry with Annexin-V/PI,and the cell cycle was analyzed by FACS.Results Compared with the use of As2O3 seperately,the combinatrion of AS2O3 and VitC could increase the inhibition ability and the apoptosis rate of Hep-2 cells markedly(P<0.05).Combination effectively enhance the change of cell cycle by As2O3;enhance cell cycle's arresting in G2 and M phase,and so enhance cell's apoptosis.Conclusion Combination of As2O3with VitC could increase apoptosis rate of Hep-2 cells markedly,the effect may be related with its influence on cell cycle.
