黄芪甲苷对肾间质纤维化的拮抗作用

摘    要:目的:探讨黄芪甲苷对大鼠单侧输尿管梗阻模型(UUO)肾间质纤维化的影响。方法:27只雄性SD大鼠随机分为假手术组、模型组、黄芪甲苷组,每组9只。于治疗2周后,心脏采血,检测血肌酐、尿素氮;取梗阻侧肾组织,免疫组织化学染色观察TGF-β、CTGF、FN的阳性表达,并进行半定量分析。结果:与假手术组相比,模型组血肌酐、尿素氮的水平以及TGF-β、CTGF、FN的表达均显著增高。与模型组相比,黄芪甲苷组血肌酐、尿素氮水平降低,TGF-β、CTGF、FN的阳性表达显著下调。结论:黄芪甲苷可能通过降低大鼠血肌酐、尿素氮,抑制TGF-β、CTGF、FN的表达,改善肾功能作用,从而延缓肾间质纤维化病程的进展。

Abstract:Objective : To explore the effects of Astragalus Saponin I ( ASI ) on renal interstitial fibrosis following unilateral ureteral obstruction (UUO) in rats. Methods:27 male SD rats were randomly divided into 3 groups ,9 in each group:the sham operated group, UUO group and AS[ group. After treatment for additional 14 days ,the rats were killed to get their blood through heart. The levels of Scr and BUN were measured and the semiquantitative assessments of interstitial fibrosis degree and immunohisto- chemical studies localizing Transforming growth factor - β ( TGF - β ) , Connective tissue growth factor (CTGF) and Fibronectin (FN) were carried out. Results:The levels of Scr and BUN and the expressions of TGF - β, CTGF and FN in the model group were significantly higher than those in the sham operated group. Compared with model group, the levels of Scr and BUN and the expressions of TGF-β, CTGF and FN in the ASI group were obviously decreased. Conclusion:ASI alleviates renal interstitial fibrosis possibly via down regulating the levels of Scr and BUN and the expressions of TGF - β, CTGF and FN to improve the renal function .
