摘    要:目的探讨野生型人IL-13(whIL-13)及其变异体(mhIL-13,Arg130Gln)对培养的气道平滑肌细胞(AM-SCs)信号传导的影响。方法利用不同浓度的whIL-13及mhIL-13分别对AMSCs进行刺激,收集细胞,流式细胞仪检测STAT-6磷酸化情况,从而判断这两种蛋白分子在信号传导上的差异。结果whIL-13及mhIL-13引起的STAT-6磷酸化随浓度增加而增强。mhIL-13在较低浓度(0.3 ng/ml)即可引起STAT-6磷酸化,而whIL-13则需要更高浓度(1 ng/ml);相同浓度时STAT-6磷酸化情况在两种蛋白分子间具有显著性差异;mhIL-13作用强度大约为whIL-13分子的3-5倍。结论mhIL-13分子致气道平滑肌细胞STAT-6磷酸化能力明显强于whIL-13,低浓度mhIL-13即可在体外培养的AMSCs中引起信号传导。

Abstract:Objective To investigate the effects of wild-type human IL-13(whIL-13) and its mutant(mhIL-13,Arg130Gln) for signaling on airway smooth muscle cells(AMSCs) cultured in vitro.Methods Simulateing AMSCs with whIL-13 and mhIL-13 in different concentrations,and then collected these cells.Detection of phosphorylated STAT-6 were done by flow cytometry,Compare the difference between these two kinds of protein.Results Phosphorylations of STAT-6 stimulated by whIL13 and mhIL-13 respectively were enhance accompany with cytokine concentrations.mhIL-13 can induce phosphorylation of STAT-6 in lower concentration(0.3 ng/ml),while whIL-13 need higher concentration(1 ng/ml).Phosphorylations of STAT-6 between whIL-13 and mhIL-13 in the same concentration show significant difference.Effect intensity of mhIL-13 is 3-5 times approximately than whIL-13.Conclusion The potentia of mhIL-13(Arg130Gln) to induce phosphorylation of STAT-6 on AMSCs shows stronger obviously than whIL-13.mhIL-13 can signaling on AMSCs with a lower concentration.
