三氧化二砷白蛋白微球抑制人膀胱癌细胞生长的实验研究

摘    要:目的:研究三氧化二砷白蛋白微球(AsO3—HAS—NS)对人膀胱癌细胞生长的抑制作用。方法:用交联固化的方法制备As2O3—HAS—NS,用原子吸光光度法测量微球中的砷浓度,采用显微镜观察和MTT法研究As2O3—HAS—NS对细胞生长的抑制作用,用^3氢—胸腺嘧啶核苷(^3H—TDR)掺人法测定其体外杀伤活性。用流式细胞术(FCM)检测细胞凋亡。结果:显微镜观察及MTT法显示As2O3—HAS—NS对人膀胱癌EJ细胞体外生长有明显抑制作用,呈现出时间及剂量依赖性。^3H—TDR掺人法测得As2O3—HAS—NS作用的EJ细胞存活率开始较高,但随着时间的延长,较As2O3降低快。流式细胞分析显示As2O3—HAS—NS有明显地促进EJ细胞凋亡的作用,Gn/G1期百分比明显减少,而G2/M期百分比明显增多,至24h达57.1%。24h Sub—G1期细胞占9.4%。结论:As2O3—HAS—NS对EJ细胞的作用优于单纯As2O3,可能成为实体肿瘤治疗的更优剂型。

Abstract:Objective:To study the effects of As 2O 3-HAS-NS on the growth of human bladder cancer cell.Methods:As 2O 3-HAS-NS were prepared by using methods of chemical cross-linking,the concentration of As 2O 3 was determined by atomic fluometry method,the growth inhibition of tumor cell was studied by the microscope observation and MTT method. 3H-TDR incorportion method was used to show killing activity in vitro,and cell apoptosis was observed by FCM.Results:As 2O 3-HAS-NS could inhibit the growth of EJ cell effectively,which depended on the effect time and concentration of drug. 3H-TDR incorportion method showed survival rate of EJ cell was high(86.16%) after As 2O 3-HAS-NS was used for 0.5 h,but As 2O 3-HAS-NS caused survival rate to decrease more quickly than As 2O 3.FCM analysis showed As 2O 3-HAS-NS could promote EJ cell apoptosis.Cell cycles analysis showed G 0/G 1 phase cells percentage decreased and G 2/M phase percentage increased remarkably,which showed that apoptosis cell were blocked in G 2/M phase. Sub-G 1 phase cells percentage was 9.4% at 24 h.Conclusions:As 2O 3-HAS-NS showed higher activity of killing bladder tumor cells than As 2O 3 and might be a kind of better pharmaceutics to treat stereo-tumor.
