谷氨酰胺对肠粘膜上皮细胞合成多聚免疫球蛋白受体的影响

摘    要:目的:研究谷氨酰胺对体外培养肠上皮细胞合成多聚免疫球蛋白受体的胞外部分分泌片的影响,以探讨谷氨酰胺保护肠粘膜屏障功能的作用机制。方法:Caco-2细胞(20-30代)应用含有不同浓度谷氨酰胺(0、0.1、1、4mM)的DMEM培养液培养7d,细胞生长达到融合后进行实验。应用蛋白印迹杂交方法检测各组分泌片蛋白的合成;应用实时定量PCR方法检测各组分泌片mRNA的表达。结果:Western blot分析显示随着谷氨酰胺浓度的增加,Caco-2细胞合成分泌片蛋白的含量明显增加。实时定量PCR结果与Western blot结果一致,随着谷氨酰胺浓度的增加,Caco-2细胞分泌片mRNA的表达逐渐升高。结论:谷氨酰胺可以促进Caco-2细胞分泌片蛋白的合成,这一作用通过增加分泌片mRNA表达实现。

Abstract:Objective:This study detected the effect of glutamine(GLN) on the expression of secretory component(SC) in Caco-2 cell in order to explore the mechanisms of glutamine(GLN) in maintenance of intestinal barrier function.Methods:Caco-2 cells between passage 20 and 30 were cultured with indicated concentration of GLN DMEM(0、0.1、1、4 mM) for 7 days before treatment.The expression of SC protein was detected by immunoblotting.Real time RT-PCR was used to detect the expression of secretory component SC mRNA.Results:Glutamine caused a concentration dependent increase of SC protein in Caco-2 cells by Western blot analysis.The result of real time RT-PCR was similar with that of Western blot analysis that glutamine caused a concentration dependent increase of SC mRNA in Caco-2 cells.Conclusions: Glutamine increased the expression of SC protein in Caco-2 cell which is due to the elevation of SC mRNA.
