益气活血复方对兔颈总动脉球囊损伤后PDGF-B mRNA表达的影响

目的观察益气活血复方对兔颈总动脉内皮损伤后血小板源性生长因子-B(PDGF-B)mRNA表达的影响。方法将雄性新西兰白兔28只随机分为3组:空白组给予普通饲料喂养8周;模型组和益气活血复方组给予高脂饲料喂养6周后,用球囊拉伤法建立兔颈总动脉内皮损伤模型。第8周末处死动物,用RT-PCR法检测颈动脉损伤处PDGF-B mRNA的表达,HE染色观察颈动脉组织病理形态学改变。结果与模型组对比,益气活血复方可明显抑制动脉损伤后的内膜增殖反应,下调损伤处PDGF-B mRNA的表达水平(P<0.05)。结论益气活血复方可以显著减轻兔血管成形术后内膜增生,其机制可能与抑制PDGF-B mRNA表达有关。

Effect of Yiqi Huoxue compound decoction on the expression of PDGF-B mRNA in Rabbits after saccule injury of carotid artery

Objective It is to observe the effects of Yiqi Huoxue compound decoction(YHCD) on the expression of platelet-derived growth factor(PDGF)in rabbits after injury of common carotid artery endothelium.Methods 28 male New Zealand white rabbits were randomly divided into three groups.The control group was fed with common animal feeds for 8 weeks,the model group and the YHCD group were fed with high-fat feeds for 6 weeks,then their saccule of carotid artery was damaged to establish animal model.After 8 weeks,all the animals were killed to determine PDGF-B mRNA in injury part of carotid artery with RT-PCR,and to observe the pathologic change of carotid artery tissue with HE staining.Results Compared with model group,YHCD could obviously inhibit proliferative response of endomembrane after artery injury,and low down the expression level of PDGF-B mRNA(P<0.05).Conclusion YHCD obviously lighten the intimal hyperplasia after angioplasty in rabbits,and the mechanism may be related with inhibiting the expression of PDGF-B mRNA.
