大蒜素对金属基质蛋白酶表达的影响

摘    要:目的:研究大蒜素对高脂血症家兔金属基质蛋白酶(MMP-13)表达的影响。方法:选用健康新西兰雄性大白兔36只,体重2.24-0.3。随机将36只新西兰雄性大白兔分为三组:(1)空白对照组(正常组)12只,每天喂食普通饲料;(2)阳性对照组(模型组)12只,每天喂食1g胆固醇+10%猪油+普通饲料;(3)大蒜素组(治疗组)12只,每天喂食1g胆固醇+10%猪油+普通饲料+8mg/kg大蒜素。实验开始时,每天7:00治疗组腹腔注射大蒜素,其他两组腹腔注射等量的生理盐水2.5mg/kg。在第16周末,测量血脂指标(TC、TG、HDL—C、LDL—c)以及脂质过氧化产物丙二醛(MDA)、超氧化物歧化酶(SOD)的指标后,处死全部家兔。取主动脉标本观察并记录其形态学结果后进行HE染色观察并记录标本的病理变化,通过免疫组织化学方法观察并记录标本斑块内金属基质蛋白酶(MMP-13)的表达情况。结果:大蒜素能够降低血中TC、TG、LDL—C、升高HDL—C以及减少丙二醛的含量,并提高超氧化物歧化酶SOD的活力,同时能够降低斑块中金属基质蛋白酶(MMP-13)的阳性表达率。减小已经形成的动脉粥样硬化斑块的作用。结论:大蒜素具有降低血脂,抗脂质过氧化,降低斑块中金属基质蛋白酶(MMP-13)的阳性表达率,预防动脉粥样硬化形成的作用外,还对动脉粥样硬化斑块的消退有一定的影响。

Abstract:Objective:To investigate the effect of garlicin on the expression of MMP - 13 level in sample atherosclerosis (AS) plaques. Methods:Thirty six healthy male New Zealand white rabbits (weight :2.2 ± 01. 3kg) were randomly divided into 3 groups, 12 rabbits in every group : ( 1 ) 览 control group ( normal group), fed with general feed every day, ( 2 ) positive control group ( model group), fed with 1 g cholesterol, 10% lard and general feed, (3)Garlicin group( prophylaxis group), fed with 1 g cholesterol, 10% lard,8 mg/ kg garlicin and general feed. From the beginning of the experiment, positive control group was given 2.5 mg/kg garlicin celiac injection,and other groups were given 2.5 mg/kg saline water celiac injection every day. At the end of 16 weeks,the levels of serum TC , TG , LDL - C , HDL - C , MDA and SOD were measured. Then all rabbits were kill and the MMP - 13 level in sample atherosclerosis plaques were studied by means of immunohistochemistry. Results: It was found that garlicin could decrease the levels of the serum TC , TG , LDL - C and MDA in garlicin group compared with those in positive control group,increase the serum HDL - C and SOD concentration, decrease the rate of positive expression of MMP- 13 level in sample AS plaques and alleviate the atherosclerotic lesion. Conclusion: garlicin can cure the formatted atherosclerosis plaques besides lowering serum lipids, decreasing the peroxidation of lipids and the rate of positive expression of MMP - 13 level in sample AS plaques and preventing the formation of atherosclerosis.
